The large-caliber primary afferents innervating the spindles of jaw-closing muscles have their cell bodies located centrally in mesencephalic trigeminal nucleus (NVmes). We shown, an acid-induced jaw muscle chronic myalgia model, that these exhibit increased excitability and ectopic discharges emerge from subthreshold membrane oscillations (SMOs) supported by a persistent sodium current (I NaP) exquisitely sensitive to extracellular Ca2+-decreases. Here, we explore if Ca2+-binding astrocytic protein, S100β, contributes this hyperexcitability emergence aim localize site where discharge arises using whole-cell patch-clamp recordings on mice brain slices. found astrocytes, lowering [Ca2+]e at focal points along axons NVmes neurons through enhance amplitude NaV1.6-dependent SMOs, leading firing. Abstract Facial expressions and movements, from a subtle ephemeral grimace, to vigorous rapid chewing, offer direct insights into the moment-to-moment change of neural physiological processes. Mice, with discernible facial responses evolutionarily conserved mammalian movement control circuits, provide an ideal model unravel link between underlying states. However, existing frameworks lack spatial or temporal resolution sensitively track all movements mouse face, due its small conical form factor. We introduce Cheese3D, computer vision system that captures high-speed 3D motion entire face (including ears, eyes, whisker pad, jaw, while covering both sides face) using calibrated six-camera array. The interpretable framework extracts dynamics anatomically-meaningful features in absolute world units at sub-millimeter precision. precise face-wide data generated by Cheese3D provides clear insights, as shown proof-of-principle experiments predicting time under general anesthesia changing patterns, inferring tooth muscle anatomy fast ingestion motions across measuring minute differences evoked brainstem stimulation, relating activity spontaneous including expressive only measurable (e.g., angles ear motion). can serve discovery tool renders highly readout otherwise hidden internal

ABSTRACT Background Chewing is a fundamental motor activity, but there no specific assessment tool in Italian for paediatric rehabilitation. The Karaduman Performance Scale (KCPS) performance‐based that allow to classify chewing performance childhood. Objective To translate, culturally adapt and assess reliability, criterion validity cross‐cultural of the KCPS into population. Methods Following international guidelines, was translated adapted Italian. Inter‐rater reliability measured using intraclass correlation coefficient (ICC), Pearson comparing score with Paediatric Screening‐Priority Evaluation Dysphagia (PS‐PED), examined across diagnostic groups. Results study included 165 children mean age 6.33 different health conditions, namely autism spectrum disorder, cerebral palsy genetic syndromes. analysis revealed reliable measure ICC 0.93, moderate positive linear PS‐PED (Pearson 0.48) found. In each group, disorders were found, highlighting characteristics. Conclusions Despite limited sample need exploring relationship abilities severity diseases, found valid determining level Now clinicians can use it more confidence their clinical practice research.

Abstract The first phase of feeding consists in acquiring solid foods from the environment by biting, and their preparation for swallowing chewing. These actions require precise coordination tens orofacial muscles jaw tongue. siege this motor patterning is known to be reticular formation, a complex poorly mapped region hindbrain, but neuron groups involved are still elusive. Here, we characterize group interneurons located supratrigeminal area that express homeodomain transcription factor Phox2b . This nucleus — Sup5 premotor both jaw-closing jaw-opener motoneurons receives direct input cranial sensory afferents, cortex satiation related nuclei. Its activity differentially tracks lapping, biting chewing movements, suggesting its involvement elaboration distinct patterns vivo. Acute global activation or inhibition optogenetics interrupt volitional sequences. Thus, an obligatory subcortical node, topologically genetically defined, neural circuits control oral feeding.
