Gas fermentation offers a carbon-neutral route for producing industrial feedstocks using autotrophic microbes to convert carbon dioxide (CO2) in waste gases, such as emissions and biogas, into valuable chemicals or biofuels. However, slow microbial metabolism owing low gaseous solubility causes significant challenges gas fermentation. Although chemical genetic manipulations have been explored improve fermentation, they are either nonsustainable complex. Herein, an artificial soil-like material (SLM) inspired by natural soil was fabricated the growth ofCupriavidus necatorfor enhanced poly-β-hydroxybutyrate (PHB) biosynthesis from CO2 hydrogen (H2). Porous SLM comprises low-cost nanoclay, boehmite, starch serves biocarrier facilitate colonization of bacteria delivery bacteria. With 3.0 g/L addition, water increased ∼4 times biomass PHB production boosted 29 102%, respectively, 24 h culture. In positive modulation observed biosynthesis. biosynthesis-associated gene expression found be response addition. The concentrations intermediates metabolic pathway biosynthesis, pyruvate acetyl-CoA, well reducing energy (ATP NADPH) significantly with also demonstrated merits easy fabrication, high stability, recyclability, plasticity, thereby indicating its considerable potential large-scale application

The transcriptional architecture of phages can deepen our understanding the phage-host infection process and be key importance for phage engineering biotechnological applications. Here, we applied ONT-cappable-sequencing, a long-read RNA-sequencing technique, to study regulatory mechanisms Pseudomonas infecting giant 201ϕ2-1. We identified 67 promoters 132 terminators that together represent 92 units. A full comparison these data transcriptome model ϕKZ confirmed programs prototypes Serwervirus Phikzvirus genera are largely conserved, despite some subtle differences. Evidence supporting shared include identification highly similar sequence motifs elements in both conservation loci relative homologous genes each phage. Moreover, discovered sRNA 201ϕ2-1 is conserved among prototype members different genera. Sequencing host genome resulted its reclassification as atacamensis, close important agricultural biocontrol agent chlororaphis. Finally, conducted vivo assays eight found them strongly terminate transcription P. Control from have rich history applications biotechnology. In studies, demonstrate new insight into program potential novel useful tools synthetic biology circuitry.
