The German Environmental Specimen Bank (ESB) annually archives 24-h urine samples since the early 1980s. In this study, we analyzed 420 of these from years 2014 to 2022 for metabolites 18 phthalates and two substitutes. We merged new data with previous measurement campaigns a combined dataset 1825 covering 35-year period 1988 investigate time trends, calculate daily intakes perform an anti-androgenic mixture risk assessment. With extended set 41 biomarkers, are now able monitor exposure all EU-labelled reprotoxic phthalates. Most phthalate exposures continued decrease first measurements in 80s, biggest drops DnBP (96.6 %) DEHP (90.9 %). DiNP DiDP, seen on rise earlier campaigns, declined. Exposures newly included, were generally negligible. Regarding risk, 5 % highly exposed still exceeded Hazard Index (HI) 1 2009. current campaign only three individuals (0.7 HI (with exceedances driven by DnBP).In 2022, 20 had > 0.2, which propose as benchmark interpreting considering concurrent other anti-androgens. Exposure substitutes DINCH DEHTP continues increase, exceeding those 2018. Compared United States (US) National Health Nutrition Examination Survey (NHANES) seem align, except up ten times higher levels US. Human biomonitoring (HBM) is ideal tool capture actual per individual, integrating external sources pathways, thus will continue use HBM assessment (anti-androgenic) chemicals.

Consumer product chemicals pose an environmental risk to public health. Exposure during pregnancy consumer chemicals, particularly phthalates and phenols, may increase the susceptibility disorders by dysregulating inflammation oxidative stress. However, existing studies rely on downstream nonmodifiable markers of these processes. Oxylipins are oxidized lipids that act as key upstream drivers Importantly, oxylipins responsive therapeutic interventions thus potentially modifiable. Using recent advances in lipidomics statistical approaches address both individual chemical biomarkers their mixtures, we determined associations between early exposure a large prospective cohort. Overall, our results revealed among produced across several biosynthetic pathways, suggesting pattern indicative dysregulated elevated levels Phthalate metabolites were primary associations, for low molecular weight phthalates, often used personal care products. found similar biomarker phthalate replacement is increasingly Our study provides observational evidence specific physiological pathways be including legacy replacements.

Inflammation and oxidative stress are critical to pregnancy, but most human study has focused on downstream, non-causal indicators. Oxylipins lipid mediators of inflammation that act through many biological pathways. Our aim was characterize predictors circulating oxylipin concentrations based maternal characteristics.
