Anogenital distance (AGD) is a postnatal marker of in utero exposure to androgens and anti-androgens, predictor reproductive health. Pesticides, which are associated with endocrine dysfunction, immunological dysregulation, and cancer, widespread sources of drinking water contamination. The state Paraná has a population 11 million, is the second largest grain producer in Brazil leading consumer pesticides. In this study, we analyzed extent contamination from proven, probable, or potentially carcinogenic pesticides (alachlor, aldrin-dieldrin, atrazine, chlordane, DDT-DDD-DDE, diuron, glyphosate-AMPA, lindane-γ-HCH, mancozeb-ETU, molinate, trifluralin) 127 grain-producing municipalities Paraná. Abstract Background Herbicides are environmental contaminants that have gained much attention due to the potential hazards they pose human health. Glyphosate, active ingredient in many commercial herbicides, is most heavily applied herbicide worldwide. Poly- and perfluoroalkyl substances (PFAS) are ubiquitous persistent environmental contaminants that may act as endocrine disruptors in utero, but the specific pathways unknown. We examined associations between maternal serum PFAS sex steroid hormones at three time points during pregnancy. Pregnant women participating Understanding Pregnancy Signals Infant Development (UPSIDE) study contributed biospecimens, questionnaire, medical record data each trimester (n = 285). (including perfluorooctanoic acid (PFOA), perfluorooctanesulfonic (PFOS), perfluorohexane sulfonic (PFHxS), perfluorononanoic (PFNA) perfluorodecanoic (PFDA)) were analyzed second-trimester samples by high-performance liquid chromatography tandem mass spectrometry (LC-MS/MS). Total testosterone [TT], free [fT], estrone [E1], estradiol [E2], estriol [E3]) measured LC-MS/MS from trimester. Linear mixed models with random intercepts used to examine log-transformed concentrations hormone levels, adjusting for covariates, stratifying fetal sex. Results presented mean percentage difference (Δ%) levels per ln-unit increase concentration. In adjusted models, PFHxS was associated higher TT (%Δ 20.0, 95%CI: 1.7, 41.6), particularly among carrying male fetuses 15.3, 1.2, 30.7); this association strengthened pregnancy progressed.
