Background: Pain is a major clinical and socioeconomic problem worldwide. The available therapies are not always effective often associated with the multiple adverse effects that reduce their application. Natural compounds an important group of pharmaceuticals may be used in pain management. We aimed to investigate analgesic activity sanguinarine–chelerythrine from Coptis chinensis. Methods: anti-inflammatory fraction C. chinensis extract (SC 5 10 mg/kg), sanguinarine (SAN 1 2 mg/kg) chelerythrine (CHEL 4 8 was assessed tail flick formalin tests. Adiponectin, a hormone secreted by adipocytes, has anti-inflammatory effects and is involved in various physiological pathological processes such as obesity, inflammatory diseases, cartilage diseases. However, the function of adiponectin intervertebral disc (IVD) degeneration not well understood. This study aimed to elucidate AdipoRon, an agonist receptor, on human IVD nucleus pulposus (NP) cells, using three-dimensional vitro culturing system. also AdipoRon rat tail tissues vivo puncture-induced model. Analysis quantitative polymerase chain reaction demonstrated downregulation gene expression proinflammatory catabolic factors interleukin (IL)-1β (10 ng/mL) NP cells treated with (2 μM). Furthermore, western blotting showed AdipoRon-induced suppression p65 phosphorylation (p < 0.01) under IL-1β stimulation adenosine monophosphate-activated protein kinase (AMPK) pathway. Intradiscal administration was effective alleviating radiologic height loss induced annular puncture IVD, histomorphological degeneration, production extracellular matrix factors, cytokines.
