Tetrabromobisphenol A (TBBPA) is a common brominated flame retardant that has wide range of toxic effects on organisms. However, the mechanism TBBPA digestive system rarely been studied. The purpose this study was to investigate toxicity gastric mucosa. In study, (mixed with corn oil) administered by gavage at doses 0 mg/kg (CG), 10 and 20 mg/kg. results showed levels ROS, MDA LPO were increased, activities antioxidant enzymes decreased. Large amounts ROS activated NF-κB pathway, leading development an inflammatory response. expression BCL family Caspase (Cas) genes inducing apoptosis. RIP3/MLKL pathway activated, cell necrosis. summary, can cause damage mucosa through oxidative stress, increased activation pathway. Treatment NAC alleviated caused TBBPA.

The host-bacterial interactions play the key role in inflammatory bowel disease (IBD). Dysbiosis of intestinal flora can lead to pathological changes intestine. Rosmarinic acid (RA) is a natural phenolic compound with antioxidant, anti-cancer, anti-inflammatory, anti-apoptotic, anti-fibrotic, and anti-bacterial activities that has palliative effect on acute IBD. We have established an vivo model for mice. Histological staining was performed directly observe RA alterations tract. alteration mouse observed by 16S rRNA high-throughput sequencing, mechanism action detected qPCR western blot. results showed had significant protective upregulated abundance Lactobacillus johnsonii Candidatus Arthromitus sp SFB-mouse-NL downregulated Bifidobacterium pseudolongum, Escherichia coli, Romboutsia ilealis. expressions ROCK, RhoA, CaM, MLC, MLCK, ZEB1, ZO-1, ZO-2, occludin, E-cadherin, IL-1β, IL-6, TNF-α, GRP78, PERK, IRE1, ATF6, CHOP, Caspase12, Caspase9, Caspase3, Bax, Cytc, RIPK1, RIPK3, MLKL, expression IL-10 Bcl-2. These displayed inhibited inflammation, which caused tight junction damage, repairing dysbiosis, relieved endoplasmic reticulum stress, cell death, corrected smooth muscle contractile dysregulation. this study revealed could small intestine mice regulating flora. IMPORTANCE Inflammatory (IBD) chronic, relapsing, remitting disorder gastrointestinal system. In study, we investigated effects rosmarinic effective reducing contraction abnormalities, disorders.

ABSTRACT Given the burgeoning Nyctereutes procyonoides breeding industry and its growing scale, it is imperative to investigate impact of high-fat diets on health these animals. This study involved 30 male comparable weights (3 kg ±0.5), randomly assigned either a control group or diet ( n = 15 each). The latter was fed mixture lard basal in 2:5 ratio, establishing model procyonoides. induced diarrhea histopathological changes Analysis small intestine contents using 16S rRNA sequencing revealed diet-induced disruption gut microbiota. Specifically, Escherichia-Shigella emerged as biomarker P 0.049), while Vagococcus prevalent indicating significant increase harmful bacteria group. Furthermore, this disrupted flora correlated with inflammation oxidative stress, evidenced by marked increases TNF-α < 0.01), IL-1β 0.05), IL-6 0.05) levels, measured via q-PCR, Western blot, stress assays. In addition, q-PCR analysis upregulation apoptosis necrosis markers, including Bax, Caspase3, Caspase9, Caspase12, RIPK3, RIPK1 0.01 0.001), concurrent downregulation anti-apoptotic gene Bcl-2 0.01) group, consistent protein expression trends. These findings suggest that alters microbiome toward more bacterial composition, escalating inflammatory responses intestinal tissue permeability, culminating cell necrosis. IMPORTANCE examines Our research established diet, revealing impacts, such diarrhea, histological anomalies, alterations emphasize importance preventing issues promoting sustainable growth. They highlight microbiota overall animal health.
