The constant appearance of new severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) variants concern (VoCs) has jeopardized the protective capacity approved vaccines against disease-19 (COVID-19). For this reason, generation vaccine candidates adapted to emerging VoCs is special importance. Here, we developed an optimized COVID-19 candidate using modified vaccinia virus Ankara (MVA) vector express a full-length prefusion-stabilized SARS-CoV-2 spike (S) protein, containing 3 proline (3P) substitutions in S protein derived from beta (B.1.351) variant, termed MVA-S(3Pbeta). Preclinical evaluation MVA-S(3Pbeta) head-to-head comparison previously generated MVA-S(3P) candidate, expressing Wuhan (with also 3P substitutions), demonstrated that two intramuscular doses both fully protected transgenic K18-hACE2 mice lethal challenge with reducing mRNA and infectious viral loads lungs bronchoalveolar lavages, decreasing lung histopathological lesions levels proinflammatory cytokines lungs. Vaccination elicited high titers anti-S Th1-biased IgGs neutralizing antibodies ancestral strain alpha, beta, gamma, delta, omicron. In addition, similar systemic local S-specific CD4 + CD8 T-cell immune responses were by after single intranasal immunization C57BL/6 mice. These preclinical data support clinical MVA-S(3P), explore whether they can diversify potentially increase recognition protection VoCs.

Bacterial infections have attracted the attention of researchers in recent decades, especially due to special problems they faced, such as their increasing diversity and resistance antibiotic treatment. The emergence development SARS-CoV-2 infection stimulated even more research find new structures with antimicrobial antiviral properties. Among heterocyclic compounds remarkable therapeutic properties, benzimidazoles, triazoles stand out, possessing antimicrobial, antiviral, antitumor, anti-Alzheimer, anti-inflammatory, analgesic, antidiabetic, or anti-ulcer activities. In addition, literature last decade reports benzimidazole-triazole hybrids improved biological properties compared simple mono-heterocyclic compounds. This review aims provide an update on synthesis methods these hybrids, along activities, well structure-activity relationship reported literature. It was found that presence certain groups grafted onto benzimidazole and/or triazole nuclei (-F, -Cl, -Br, -CF3, -NO2, -CN, -CHO, -OH, OCH3, COOCH3), some heterocycles (pyridine, pyrimidine, thiazole, indole, isoxazole, thiadiazole, coumarin) increases activity hybrids. Also, oxygen sulfur atom bridge connecting rings generally mentions only benzimidazole-1,2,3-triazole Both for additional ring activity, which is agreement three-dimensional binding mode summarizes advances derivatives potential agents covering articles published from 2000 2023.

Abstract The global circulation of SARS-CoV-2 has been extensively documented; however, the dynamics within Central America, particularly Nicaragua, remain underexplored. This study characterizes genomic diversity in Nicaragua from March 2020 through December 2022, utilizing 1064 genomes obtained via next-generation sequencing. These sequences were selected nationwide and analyzed for variant classification, lineage predominance, phylogenetic diversity. We employed both Illumina Oxford Nanopore Technologies all sequencing procedures. Results indicated a temporal spatial shift dominant lineages, initially B.1 A.2 early to various Omicron subvariants toward study’s end. Significant shifts correlated with changes COVID-19 positivity rates, underscoring epidemiological impact dissemination. Comparative analysis regional data underscored low circulating lineages their delayed introduction compared other countries American region. also linked specific viral mutations hospitalization emphasizing clinical relevance surveillance. research advances understanding evolution provides valuable information regarding its genetic public health officials America. highlight critical role ongoing surveillance identifying emergent informing strategies.

The recently discovered coronavirus, known as SARS-CoV-2, is a highly contagious and potentially lethal viral infection that was declared pandemic by the World Health Organization on March 11, 2020. Since beginning of pandemic, an unprecedented number COVID-19 vaccine candidates have been investigated for their potential to manage pandemic. Herein, we reviewed development associated research effort, both traditional forward-looking, demonstrate advantages disadvantages technology, in addition efficacy limitations against mutant SARS-CoV-2. Moreover, report repurposed drug discovery, which mainly focuses virus-based host-based targets, well inhibitors. SARS-CoV-2 targets include main protease (Mpro), RNA-dependent RNA-polymerase (RdRp), are most well-studied conserved across coronaviruses, enabling broad-spectrum inhibitors these enzymes.
