Abstract Purpose Oral administration of the first-line antidepressant paroxetine (PAX) has certain limitations, including difficulty in reaching brain due to oral absorption. Although there are many invasive and non-invasive strategies available cross blood–brain barrier (BBB), they run counter long-term convenience for patients. Methods We herein designed a simple PAX-loaded nanoparticle-integrated dissolving microneedles system (PAX-NP-DMNs), aiming improve bioavailability PAX through synergistic permeation-enhancing effect (MNs) nanoparticles (NPs). Results assessed NPs characteristics before after MNs preparation confirmed successful construction PAX-NP-DMNs based on differential scanning calorimetry, X-ray diffraction, Fourier transform infrared spectroscopy. In mechanical strength test, addition increased by 0.43 times prolonged release from 1 h 48 h; over 40 than solution isolated skin penetration permeability experiments. Microneedles (MNs) prepared from polymeric materials are painless and minimally invasive, safe efficient, but they hindered by low mechanical strength single diverse drug release pattern. Due to the distinctive dimensions of poly(lactic-co-glycolic acid) (PLGA) nanoparticles (NPs), integration nano-technology with microneedles can effectively improve penetration delivery efficiency through stratum corneum. We herein designed a simple paroxetine (PAX)-loaded PLGA nanoparticles-integrated dissolving system (PAX-NPs-DMNs), aiming bioavailability PAX synergistic permeation-enhancing effect (DMNs) NPs. PAX-NPs-DMNs had complete tips molding rate (Neff) (94.06 ± 2.16) %, 15×15 quadrangular-conical microneedle array an overall fracture force 301.10 N, which were improved nearly 0.50 times compared blank (HA-DMNs) (PAX-DMNs). could extend duration 1 h 24 cumulative permeability per unit area (Qn) was 47.66 7.37 higher than PAX-DMNs groups. be rapidly dissolved within 10 min without hindering skin healing or causing adverse reactions. This study confirmed that DMNs, easily penetrate provide sustained effects, thus offering more convenient effective method for central nervous diseases.
