Abstract Dietary polysaccharides have the function of regulating gut microbiota and metabolites, can serve as an important source functional foods. This study aimed to analyze regulation poria cocos on structure metabolism human through in vitro fecal fermentation model. The results showed that Poria be utilized by microbiota, resulting in a significant decrease pH increase concentration short-chain fatty acids. Additionally, relative abundance beneficial bacteria such Lactobacillus, Bifidobacterium increased, while harmful Escherichia-Shigella, Bilophila decreased. Furthermore, had positive effect microbial significantly up-regulating metabolites L-cystine, etelcalcetide. These highlight role maintaining intestinal health indicate their potential prebiotics food industry.

Polysaccharides derived from Atractylodes chinensis (DC.) Koidz. (ACP), a traditional Chinese medicine, were extracted and analyzed for their structural characteristics anti-gouty nephropathy (GN) activity. Sprague-Dawley (SD) rats divided into six groups: control, model, positive three treatment groups (ACP-60-L, ACP-60-M, ACP-60-H). Treatment significantly reduced inflammatory responses renal damage, as evidenced by decreased levels of uric acid (UA), creatinine (Cr), blood urea nitrogen (BUN), alongside modulation NOD-like receptor protein 3 (NLRP3) expression in tissues. ACP-60 was fractionated polysaccharides, including ACP-60-A (Mw 9.18 kDa), ACP-60-B 58.21 ACP-60-C 109.01 kDa) using DEAE-52 cellulose column chromatography. Monosaccharide analysis revealed that predominantly comprised fructose (Fru) glucose (Glc), contained rhamnose (Rha), galactose (Gal), Fru, mannose (Man), included Man, Gal, Rha xylose (Xyl). In vitro studies HK-2 cells confirmed the anti-GN activity all fractions, with demonstrating highest efficacy. Structural elucidation identified its main glycosidic linkages →1)-β-Fruf-(2→ backbone α-Glcp-(1→ β-Fruf-(2→ branches. The underlying mechanism ACP-60-A’s is associated inhibition NLRP3 inflammasome signaling pathway, suppression downstream factor release, downregulation NLRP3, ASC, Caspase-1 expression. Further demonstrated superior attributable to lower molecular weight, specific monosaccharide composition, unique bond arrangement. shows potential increased efficacy through purification or modification, laying groundwork developing novel therapeutic agents GN.

Abstract Lentinan (LNT) is a polysaccharide from Shiitake mushrooms that has significant effects regarding antioxidant, hypoglycemic, anti-inflammatory activities, etc., and this effect influenced by the human intestinal flora. However, interactions between LNT microorganisms, their metabolic processes, remain unclear. In study, fermentation characteristics of its on flora metabolites were studied in vitro fermentation. Our results indicated was effectively utilised microbiota, utilisation rate 62.7 ± 3.0% after for 48 hr. The concentrations short-chain fatty acids (mainly acetic acid propionic acid) increased to 4.157 0.197 5.847 0.283 mM, respectively. also relative abundance beneficial bacteria, such as Bifidobacterium Parabacteroides, while reducing harmful bacteria like Veillonella Klebsiella. Metabolomics microbial correlation analysis revealed catabolism accompanied amino catabolism, especially alanine, aspartic acid, glutamate, biosynthesis adenosine 5’-monophosphate, etc. These findings suggest can support gut health demonstrates potential prebiotic.
