Over the past decade, Nomenclature Committee on Cell Death (NCCD) has formulated guidelines for definition and interpretation of cell death from morphological, biochemical, functional perspectives. Since field continues to expand novel mechanisms that orchestrate multiple pathways are unveiled, we propose an updated classification subroutines focusing mechanistic essential (as opposed correlative dispensable) aspects process. As provide molecularly oriented definitions terms including intrinsic apoptosis, extrinsic mitochondrial permeability transition (MPT)-driven necrosis, necroptosis, ferroptosis, pyroptosis, parthanatos, entotic death, NETotic lysosome-dependent autophagy-dependent immunogenic cellular senescence, mitotic catastrophe, discuss utility neologisms refer highly specialized instances these processes. The mission NCCD is a widely accepted nomenclature in support continued development field.

Summary Inflammasome biology is one of the most exciting and rapidly growing areas in immunology. Over past 10 years, inflammasomes have been recognized for their roles host defense against invading pathogens development cancer, auto‐inflammatory, metabolic, neurodegenerative diseases. Assembly an inflammasome complex requires cytosolic sensing pathogen‐associated molecular patterns or danger‐associated by a nucleotide‐binding domain leucine‐rich repeat receptor ( NLR ) absent melanoma 2 (AIM2)‐like receptors ALR ). s engage caspase‐1, cases requiring adapter protein apoptosis‐associated speck‐like containing CARD ASC ), to catalyze proteolytic cleavage pro‐interleukin‐1β (pro‐ IL ‐1β) pro‐ ‐18 drive pyroptosis. Recent studies indicate that caspase‐8, caspase‐11, ‐1R–associated kinases IRAK receptor‐interacting RIP contribute functions. In addition, post‐translational modifications, including ubiquitination, deubiquitination, phosphorylation, degradation control almost every aspect activities. Genetic mutations NLRP 1, 3, NLRC 4, AIM are linked with auto‐inflammatory diseases, enterocolitis, cancer. Overall, these findings transform our understanding basic clinical relevance inflammasomes. this review, we provide overview latest research discuss how activities govern health disease.
