Acute lung injury/acute respiratory distress syndrome (ALI/ARDS) is a common and serious clinical disease characterized by extensive alveolar damage inflammation leading to impaired gas exchange.Alveolar macrophages (AMs) maintain homeostatic properties immune defenses in tissues.Several studies have reported that AMs are involved regulate ALI/ARDS onset progression via different regulated cell death (RCD) programs, such as pyroptosis, apoptosis, autophagic death, necroptosis.Notably, the effects of RCD complex variable depending on environment stimuli. In this review, we provide comprehensive perspective how impacts assess its potential new therapeutic development.Additionally, describe crosstalk between types ALI, perspectives for treatment other severe diseases.

Neurodegenerative diseases (NDs) are a type of common chronic progressive disorders characterized by damage to specific cell populations in the nervous system, ultimately leading disability or death. Effective treatments for these still lacking, due limited understanding their pathogeneses, which involve multiple cellular and molecular pathways. The triggering an immune response is feature neurodegenerative disorders. A critical challenge intricate interplay between neuroinflammation, neurodegeneration, responses, not yet fully characterized. In recent years, cyclic GMP-AMP synthase (cGAS)-stimulator interferon gene (STING) pathway, crucial intracellular DNA sensing, has gradually gained attention. However, roles this pathway within types such as cells, glial neuronal its contribution ND pathogenesis, remain elucidated. review, we systematically explore how cGAS-STING signaling links various with related effector pathways under context NDs multifaceted therapeutic directions. We emphasize discovery condition-dependent heterogeneity integral diverse responses potential targets. Additionally, review pathogenic role activation Parkinson's disease, ataxia-telangiectasia, amyotrophic lateral sclerosis. focus on complex bidirectional Alzheimer's Huntington's sclerosis, revealing double-edged nature disease progression. objective elucidate pivotal pathogenesis catalyze new insights facilitating development novel strategies.

Abstract Inflammation and immune processes underlie pulmonary hypertension progression. Two main different activated phenotypes of macrophages, classically M1 macrophages alternatively M2 are both involved in inflammatory related to hypertension. Recent advances suggest that coordinate interactions among proinflammatory anti-inflammatory mediators, other cellular components such as smooth muscle cells fibroblasts. In this review, we summarize the current literature on role pathogenesis hypertension, including origin their response triggers We then discuss cytokines, vascular adventitial fibroblasts well potential therapeutic benefits disease. Identifying critical will contribute a comprehensive understanding pathophysiological abnormality, may provide new perspectives for management.

Mycobacterium tuberculosis (Mtb) is the causative pathogen of tuberculosis, most lethal infectious disease resulting in 1.3 million deaths annually. Treatments against Mtb are increasingly impaired by growing prevalence antimicrobial drug resistance, which necessitates development new antibiotics or alternative therapeutic approaches. Upon infecting host cells, predominantly macrophages, becomes critically dependent on lipids as a source nutrients. Additionally, produces numerous lipid-based virulence factors that contribute to pathogen's ability interfere with host's immune responses and create lipid rich environment for itself. As lipids, metabolism manipulating play an important role Mtb, this review provides state-of-the-art overview mycobacterial concomitant host-pathogen interaction therein. While doing so, we will emphasize unexploited bacteria-directed host-directed targets, highlight potential synergistic combinations hold promise interventions.

This study aimed to investigate the role of pyroptosis in alveolar macrophages regarding immune microenvironment acute respiratory distress syndrome (ARDS) and its prognosis.ARDS Microarray data were downloaded from Gene Expression Omnibus (GEO). Support vector machine (SVM) random forest (RF) models applied identify hub pyroptosis-related genes (PRGs) with prognostic significance ARDS. RT-PCR was used detect relative expression PRGs mRNA ARDS mice. Consensus clustering analysis conducted based on modification patterns. Bioinformatic algorithms traits biological functions Finally, protein-protein interaction (PPI) networks established regulatory proteins implications for patterns. In our study, a total 12 differential obtained. Four PRGs, including GPX4, IL6, IL18 NLRP3, identified proven be predictive ventilator-free days (VFDS) patients. The AUC values 4 0.911 (GPX4), 0.879 (IL18), 0.851 (IL6) 0.841 (NLRP3), respectively. In mice, GPX4 decreased significantly, while IL18, NLRP3 increased. Functional revealed that IL6 had strongest positive correlation CCR pathway, exhibited negative T co-inhibition pathway. Based three patterns representing different states obtained, pattern C might represent storm. The results showed plays an important finding provides new insights into pathogenesis, diagnosis, treatment
