There is a direct relationship between the duration and level of exposure to low density lipoprotein cholesterol (LDL-C) levels over one's lifespan cardiovascular events. Early treatment lower elevated LDL-C crucial for better outcomes with multiple therapies currently available reduce atherogenic lipoproteins. Statins remain foundation lowering therapy as one most cost-effective drugs atherosclerotic events (ASCVD) mortality. Nonetheless, LDL-driven goal attainment remains suboptimal globally, highlighting considerable need non-statin address residual risk related statin intolerance, non-adherence, inherited disorders. interventions beyond statins include ezetimibe, PCSK9 monoclonal antibodies, inclisiran bempedoic acid specific guideline recommendations when consider each. For patients homozygous familial hypercholesterolemia requiring more advanced therapy, lomitapide evinacumab are available, providing mechanisms that not LDL receptor dependent. Background: Lowering low-density lipoprotein cholesterol (LDL-C) is a well-established strategy for the secondary prevention of coronary heart disease (CHD). However, effectiveness specific LDL-C parameters in predicting myocardial infarction (MI) recurrence real-world settings remains inadequately explored. This study aims to examine relationship between MI and various young CHD patients. Methods: retrospective cohort involved 1013 patients aged 18–44 at time initial diagnosis, collected from cardiology department clinics Beijing Anzhen Hospital October 2022 2023. levels were assessed diagnosis final follow-up. The primary outcome was events, analyzed using survival analysis logistic regression models determine associations with parameters. Results: included (mean age: 38.5 ± 3.9 years; 94.7% men), median follow-up 1.7 years. Initially, 13.6% had <1.8 mmol/L, which increased 37.8% by study’s end. During follow-up, 96 (9.5%) experienced MI. While mmol/L baseline showed slightly lower cumulative incidence than ≥1.8 difference not statistically significant (log-rank p = 0.335). Reductions ≥50% patterns change did correlate decreased risk. <1.4 measurement associated reduced risk (adjusted odds ratio [OR]: 0.57, 95% confidence interval [CI]: 0.33–0.98) compared ≥2.6 mmol/L. Conclusions: suggests that most important parameter related history ideal target level. could potentially reduce risk, regardless levels.

Unstable atherosclerotic plaques are a major cause of acute cardiovascular events and ischemic stroke. Clinical studies have suggested link between periodontitis plaque progression, but the underlying mechanisms remain unclear. To investigate this, transcriptomic datasets related to atherosclerosis were downloaded from Gene Expression Omnibus. A weighted gene co-expression network analysis was used identify modules associated with periodontitis, Limma R package identified differentially expressed genes (DEGs) unstable stable plaques. Background Purslane ( Portulaca oleracea ) is a medicinal and edible plant. extract (POEE) exhibits lipid-lowering, anti-inflammatory, antioxidant properties. Traditionally, this has been used to treat various inflammatory conditions, including skin inflammation, enteritis, dysentery. However, its therapeutic potential molecular mechanisms in atherosclerosis (AS) remain unclear. Methods Ultra-performance liquid chromatography-quadrupole/time-of-flight mass spectrometry (UPLC-Q/TOF-MS) the Traditional Chinese Medicine Systems Pharmacology Database were employed identify active components of POEE. Network pharmacology was predict POEE’s for alleviating AS. An vitro foam cell model established by treating RAW264.7 macrophages with oxidized low-density lipoprotein (ox-LDL), protective effects POEE assessed via 3-[4,5-dimethylthiazol-2-yl]-2,5 diphenyl tetrazolium bromide (MTT) assay, while intracellular lipid accumulation identified using Oil Red O staining. Protein expression related cholesterol metabolism analyzed Western blot (WB). For vivo validation, AS induced rats through high-fat diet carotid artery injury. After 4 weeks daily administration, efficacy tested measuring serum levels, cardiac function, histopathological changes, transport-related protein expression.
