Abstract Apoptosis, a form of programmed cell death, is essential for growth and tissue homeostasis. Apoptotic bodies (ApoBDs) are extracellular vesicles (EVs) released by dying cells in the last stage apoptosis were previously regarded as debris dead cells. Recent studies unraveled that ApoBDs not but bioactive treasure left behind with an important role intercellular communications related to human health various diseases. Defective clearance infected-cells-derived possible etiology some Therefore, it necessary explore function mechanism action different physiological pathological conditions. advances have elucidated immunomodulatory, virus removal, vascular protection, regenerative, disease diagnostic potential ApoBDs. Moreover, can be used drug carriers enhancing stability, cellular uptake, targeted therapy efficacy. These reports from literature indicate hold promising diagnosis, prognosis, treatment diseases, including cancer, systemic inflammatory cardiovascular regeneration. This review summarizes recent ApoBDs-related research discusses diseases well challenges prospects ApoBDs-based therapeutic applications. Graphical

Abstract Preventing and treating avascular necrosis at the distal end of flaps are critical to surgery success, but current treatments not ideal. A recent study shows that apoptotic bodies (ABs) generated near site apoptosis can be taken up promote cell proliferation. The reveals ABs derived from fibroblast‐like cells in subcutaneous connective tissue (FSCT cells) skin promoted ischaemic flap survival. It is also found inhibited death oxidative stress M1‐to‐M2 polarization macrophages. Transcriptome sequencing protein level testing demonstrated survival endothelial macrophages by inhibiting ferroptosis via KEAP1‐Nrf2 axis. Furthermore, microRNA (miR) data vitro vivo experiments KEAP1 delivering miR‐339‐5p exert therapeutic effects. In conclusion, FSCT cell‐derived ferroptosis, macrophage transition miR‐339‐5p/KEAP1/Nrf2 axis These results provide a potential strategy administering ABs.

Metabolic reprogramming is one of the hallmarks cancer. As nutrients are scarce in tumor microenvironment (TME), cells adopt multiple metabolic adaptations to meet their growth requirements. not only present cells, but exosomal cargos mediates intercellular communication between and non-tumor TME, inducing remodeling create an outpost microvascular enrichment immune escape. Here, we highlight composition characteristics meanwhile summarize components corresponding sorting mode. Functionally, these cargos-mediated improves "soil" for metastasis. Moreover, discuss abnormal metabolism targeted by its potential antitumor therapy. In conclusion, this review updates current role TME enriches future application scenarios exosomes.

Apoptotic vesicles (ApoVs) hold great promise for inflammatory regulation and tissue repair. However, little effort has been dedicated to developing ApoV-based drug delivery platforms, while the insufficient targeting capability of ApoVs also limits their clinical applications. This work presents a platform architecture that integrates apoptosis induction, loading, functionalized proteome regulation, followed by modification, enabling creation an apoptotic vesicle system treat ischemic stroke. Briefly, α-mangostin (α-M) was utilized induce mesenchymal stem cell (MSC) being loaded onto MSC-derived as anti-oxidant anti-inflammatory agent cerebral ischemia/reperfusion injury. Matrix metalloproteinase activatable cell-penetrating peptide (MAP), microenvironment-responsive peptide, modified on surface obtain MAP-functionalized α-M-loaded ApoVs. Such engineered targeted injured brain after systemic injection achieved enhanced neuroprotective activity due synergistic effect α-M. The internal protein payloads ApoVs, upon α-M activation, were found engaged in regulating immunological response, angiogenesis, proliferation, all which contributed therapeutic effects findings provide universal framework creating systems amelioration diseases demonstrate potential neural
