The global prevalence of type 2 diabetes mellitus (T2DM) and Alzheimer’s disease (AD) is rapidly increasing, revealing a strong association between these two diseases. Currently, there are no curative medication available for the comorbidity T2DM AD. Ceramides structural components cell membrane lipids act as signal molecules regulating homeostasis. Their synthesis degradation play crucial roles in maintaining metabolic balance vivo , serving important mediators development neurodegenerative disorders. Abnormal ceramide metabolism disrupts intracellular signaling, induces oxidative stress, activates inflammatory factors, impacts glucose lipid homeostasis metabolism-related tissues like liver, skeletal muscle, adipose tissue, driving occurrence progression T2DM. connection changes levels brain, amyloid β accumulation, tau hyper-phosphorylation evident. The term type 3 diabetes mellitus (T3DM) has been considered for Alzheimer's disease (AD) due to the common molecular and cellular characteristics found between 2 (T2DM) cognitive deficits. However, specific mechanism of T3DM remains elusive, especially neuroprotective effects dietary components in hyperglycemic individuals. In this study, a peptide, Leu-Val-Arg-Leu (LVRL), walnuts significantly improved memory decline streptozotocin (STZ)- high-fat-diet (HFD)-stimulated T2DM mouse models (

The role of amyloid beta peptide (Aβ) in memory regulation has been a subject substantial interest and debate neuroscience, because both physiological clinical issues. Understanding the dual nature Aβ is crucial for developing effective treatments Alzheimer's disease (AD). Moreover, accurate detection quantification methods isoforms have tested diagnostic purposes therapeutic interventions. Abstract This study examined the effects of gingerol, an active ingredient in ginger, on monoamine oxidases (MAO)‐A and B, acetylcholinesterase (AChE), butyrylcholinesterase (BChE), α‐glycosidase enzymes as vitro. The IC 50 values were calculated for MAO‐A, MAO‐B, AChE, BChE, 13.05 µM, 16.6 2.14 nM, 3.37 27.28 respectively. Additionally, K i determined to be 0.67 ± 0.012 nM 0.86 0.033 14.36 3.41 α‐glycosidase. To evaluate antioxidant radical scavenging capacities Fe 2+ , Cu 3+ ‐TPTZ reductions, DPPH∙·and ABTS ∙+ activities examined. absorbance activity gingerol was found 1.084 0.057 reducing ability, 0.343 0.036 1.26 0.020 FRAP assay at 30 µg/mL. As results, 17.33 µg/mL DPPH∙ 5.93 assay. From these it that exhibited significant properties when compared standard antioxidants. Molecular docking simulations ADME studies have provided insights into interactions between target enzymes, guiding design novel therapeutic agents critical diseases, including Alzheimer's disease, Parkinson's diabetes.

We highlight this review neuroglial cells (astroglia, oligodendroglia, microglia) play a vital role the origin, clinical-pathological development, course brain neurodegeneration. Moreover, we include new results T2D-AD mouse model (APP+PS1 mice on high-calorie diet) investigating.
