Phthalates are extensively used as plasticizers in a variety of daily-life products, resulting widespread distribution aquatic environments. However, limited information is available on the endocrine disrupting effects phthalates organisms. The aim present study was to examine whether exposure mono-(2-ethylhexyl) phthalate (MEHP), hydrolytic metabolite di-(2-ethylhexyl) (DEHP) disrupts thyroid system fish. In this study, zebrafish (Danio rerio) embryos were exposed different concentrations MEHP (1.6, 8, 40, and 200 μg/L) from 2 h post-fertilization (hpf) 168 hpf. whole-body content hormone transcription genes involved hypothalamic-pituitary-thyroid (HPT) axis examined. Treatment with significantly decreased T4 contents increased T3 contents, indicating disruption. upregulation related metabolism (Dio2 UGT1ab) might be responsible for contents. Elevated gene Dio1 also observed which assist degrade Exposure induced development (Nkx2.1 Pax8) synthesis (TSHβ, NIS TG). encoding proteins TH transport (transthyretin, TTR) transcriptionally down-regulated after MEHP. Overall, these results demonstrate that acute alters hormones embryos/larvae changes HPT axis, thus exerting toxicity.

Di-(2-ethylhexyl) phthalate (DEHP) has the potential to disrupt thyroid endocrine system, but underlying mechanism is unknown. In this study, zebrafish (Danio rerio) embryos were exposed different concentrations of DEHP (0, 40, 100, 200, 400 μg/L) from 2 168 hours post fertilization (hpf). Thyroid hormones (THs) levels and transcriptional profiling key genes related hypothalamus-pituitary-thyroid (HPT) axis examined. The result whole-body thyroxine (T4) triiodothyronine (T3) indicated that hormone homeostasis was disrupted by in larvae. After exposure DEHP, mRNA expressions stimulating (tshβ) corticotrophin releasing (crh) increased a concentration dependent manner, respectively. expression level involved development (nkx2.1 pax8) synthesis (sodium/iodide symporter, nis, thyroglobulin, tg) also measured. transcripts nkx2.1 tg significantly after exposure, while those nis pax8 had no significant change. Down-regulation uridinediphosphate-glucuronosyl-transferase (ugt1ab) up-regulation thyronine deiodinase (dio2) might change THs levels. addition, transcript transthyretin (ttr) up-regulated, receptors (trα trβ) remained unchanged. All results demonstrated altered larvae changed HPT axis, proving induced toxicity potentially affected synthesis, regulation action hormones.

Plastic mulch films are used globally in crop production but incur considerable disposal and environmental pollution issues. Biodegradable plastic (BDMs), an alternative to polyethylene (PE)-based films, designed be tilled into the soil where they expected mineralized carbon dioxide, water microbial biomass. However, insufficient research regarding impacts of repeated incorporation BDMs on communities has partly contributed limited adoption BDMs. In this study, we evaluated effects BDM community structure function over two years geographical locations: Knoxville, TN, Mount Vernon, WA, USA. Treatments included four (three commercially available one experimental film), a biodegradable cellulose paper mulch, non-biodegradable PE no plot. Bacterial determined using 16S rRNA gene amplicon sequencing revealed significant differences by location season. Differences bacterial treatment were not for any season either location, except Fall 2015 WA observed between no-mulch plots. Extracellular enzyme assays characterize functionally, revealing sampling both TN minimal treatments. Overall, had comparable influences films.
