Polygonatum sibiricum Polysaccharide (PsP) with antioxidant function is the main active component of (P.sibiricum). The currently poor extraction yield and methods PsP cannot meet application that in food industrial production. In this research, an ultrasound-assisted extraction-deep eutectic solvents (UAE-DESs) method, which has never been used industry, was first to extract PsP. conditions were optimized by response surface method (RSM). Both simultaneously considered during optimization process. indicators PsP's level activity vitro present UAE-DESs purity, effect Under optimal conditions, included a liquid-solid ratio 26:1 (mL:g), temperature 80 °C, ultrasonic time 51 min, power 82 W, could reach (43.61 ± 0.09)%, obviously higher than single DESs (33.81%) UAE (5.83%), respectively, appeared favorably function. This research proposed efficient for PsP, filled basic gap, further improved development as dietary supplement industry.

Many chronic diseases such as Alzheimer's disease, diabetes, and cardiovascular are closely related to in vivo oxidative stress caused by excessive reactive oxygen species (ROS). Natural polysaccharides, a kind of biomacromolecule with good biocompatibility, have been widely used biomedical medicinal applications due their superior antioxidant properties. In this review, scientometric analysis the highly cited papers Web Science (WOS) database finds that activity is most studied popular among pharmacological effects natural polysaccharides. The mechanisms polysaccharides mainly contain regulation signal transduction pathways, activation enzymes, scavenging free radicals. We continuously discuss activities derivatives. At same time, we summarize field pharmaceutics/drug delivery, tissue engineering, antimicrobial food additives/packaging materials. Overall, review provides up-to-date information for further development application activities.

Polysaccharides from Polygonatum cyrtonema Hua (PSP) exert antioxidant, anti-inflammatory, and antidepressant effects. Production of reactive oxygen species (ROS) activation the calpain system NOD-like receptor protein 3 (NLRP3) inflammasome are closely related to pathogenesis depression. However, relationships among those pathways protective effects PSP have not been characterized. In this study, lipopolysaccharide (LPS) chronic unpredictable mild stress- (CUMS-) induced depression models were used evaluate mechanisms against ROS levels measured in HT-22 cells using flow cytometry. Brain tissues collected determine oxidation-related indicators inflammatory cytokines. The calpain-1, calpain-2, calpastatin, phosphatase Tensin Homolog deleted on Chromosome 10 (PTEN), suprachiasmatic nucleus circadian oscillatory (SCOP), nuclear factor-erythroid factor 2-related 2 (Nrf2), heme oxygenase-1 (HO-1), NLRP3, apoptosis-associated speck-like (ASC), caspase-1, cleaved-caspase-1, ionized calcium binding adapter molecule 1 (Iba1), phosphorylation extracellular signal-regulated kinase (p-ERK), factor-kappa B (NF-κB), interleukin-1β (IL-1β), glial fibrillary acidic (GFAP) western blotting or immunofluorescence. cellular experiments, we showed that attenuated LPS-induced production cells. animal found LPS increased expression ASC, Iba1, p-ERK, NF-κB, GFAP reduced PTEN, SCOP, Nrf2. Administration reversed these changes. N-Acetyl-L-cysteine (NAC) administration also inhibited oxidative stress NLRP3 inflammasome. Furthermore, PSP, calpeptin, MCC950 (a selective inhibitor), NAC proinflammatory cytokine release. We prevented CUMS-induced changes Nrf2 signaling depression-like behavior. These results indicate exerts through regulation stress-calpain-1-NLRP3 axis.

Polygonum sibiricum polysaccharides (PSP) are one of the main active components Polygonatum sibiricum, which is a traditional Chinese medicine with food and drug homologies. Recent studies have revealed antidepressant-like effects PSP. However, precise mechanisms not been clarified. Therefore, present study was conducted to explore that whether PSP could exert via microbiota-gut-brain (MGB) axis in chronic unpredictable mild stress (CUMS)-induced depressive mice by transplantation fecal microbiota (FMT) from administration mice. FMT markedly reversed depressive-like behaviors CUMS-induced open field, sucrose preference, tail suspension, forced swimming, novelty-suppressed feeding tests. significantly increased levels 5-hydroxytryptamine norepinephrine, decreased pro-inflammatory cytokines hippocampus reduced corticosterone, an adrenocorticotropic-hormone, serum In addition, expressions ZO-1 occludin colon lipopolysaccharide interferon-γ Moreover, regulated signaling pathways PI3K/AKT/TLR4/NF-κB ERK/CREB/BDNF. Taken together, these findings indicated exerted MGB axis.
