The present study investigated the effect of Gastrodia elata extracts (GE) on chronic electric foot-shock stress (cEFS)-induce anxiety-like behaviors in male C57BL/6 mice. This study evaluated the antidepressant effects of arecoline, a bioactive alkaloid derived from areca nuts, using mouse model depression induced by chronic unpredictable mild stress. Arecoline treatment significantly alleviated depression-like behaviors, including anxiety, anhedonia, and despair, as evidenced behavioral tests. Obesity-related cognitive dysfunction poses a significant threat to public health. The present study demonstrated mitigating effects of intermittent fasting (IF) and its combination with isomalto-oligosaccharides IF (IF + IMO) on impairments induced by high-fat-high-fructose (HFHF) diet in mice, IMO exhibiting superior effects. Transcriptomic analysis the hippocampus revealed that protective cognition might be attributed suppression toll-like receptor 4 (TLR4)/NFκB signaling, oxidative phosphorylation, neuroinflammation. Moreover, both modulated gut microbiome promoted production short-chain fatty acids, displaying more pronounced IMO-modulated microbiota, metabolites, molecular targets associated were further corroborated using human data from databases Gmrepo gutMgene. Furthermore, fecal transplantation confirmed direct impacts IMO-derived microbiota improving functions suppressing TLR4/NFκB signaling increasing BDNF levels. Notably, prior exposure prevented weight-regain-induced decline, suppressed inflammatory cytokines hippocampus, mitigated weight regain-caused dysbacteriosis without altering body weight. Our underscores IMO-augmented alleviating obesity-related impairment particularly during weight-loss weight-regain periods, presenting novel nutritional strategy tackle neurodegenerative disorders.
