Iron, an essential mineral in the body, is involved numerous physiological processes, making maintenance of iron homeostasis crucial for overall health. Both overload and deficiency can cause various disorders human diseases. Ferroptosis, a form cell death dependent on iron, characterized by extensive peroxidation lipids. Unlike other kinds classical unprogrammed death, ferroptosis primarily linked to disruptions metabolism, lipid peroxidation, antioxidant system imbalance. Ferroptosis regulated through transcription, translation, post-translational modifications, which affect cellular sensitivity ferroptosis. Over past decade or so, diseases have been as part their etiology, including cancers, metabolic disorders, autoimmune diseases, central nervous cardiovascular musculoskeletal Ferroptosis-related proteins become attractive targets many major that are currently incurable, some regulators shown therapeutic effects clinical trials although further validation potential needed. Therefore, in-depth analysis its molecular mechanisms may offer additional strategies prevention treatment. In this review, we discuss significance contribution etiology development along with evidence supporting targeting approach. Importantly, evaluate recent promising interventions, providing guidance future targeted treatment therapies against

Depression is a common mental health disorder, and in recent years, the incidence of various forms depression has been on rise. Most medications for are highly dependency-inducing can lead to relapse upon discontinuation. Therefore, novel treatment modalities therapeutic targets urgently required. Traditional Chinese medicine (TCM) offers advantages owing its multi-target, multi-dimensional approach that addresses root cause by regulating organ functions balancing Yin Yang, with minimal side effects. Cynaroside (CNS), an extract from traditional herb honeysuckle, flavonoid compound antioxidant properties. In this study, network pharmacology identified 44 potential CNS associated several correlated inflammatory signaling pathways. alleviated LPS-induced M1 polarization release factors BV-2 cells. Transcriptomic analysis validation revealed reduced polarization, lipid peroxidation, ferroptosis via IRF1/SLC7A11/GPX4 pathway. vivo experiments showed had effects similar those fluoxetine (FLX). It effectively ameliorated anxiety-, despair-, anhedonia-like states chronic unpredictable mild stress (CUMS)-induced mice microglial activation hippocampus. Thus, we conclude exerts effect inhibiting cells polarizing into phenotype reducing inflammation levels. This study provides further evidence antidepressant, offering new avenues depression.

Central neurological disorders are significant contributors to morbidity, mortality, and long-term disability globally in modern society. These encompass neurodegenerative diseases, ischemic brain traumatic injury, epilepsy, depression, more. The involved pathogenesis is notably intricate diverse. Ferroptosis neuroinflammation play pivotal roles elucidating the causes of cognitive impairment stemming from these diseases. Given concurrent occurrence ferroptosis due metabolic shifts such as iron ROS, well their critical central nervous disorders, investigation into co-regulatory mechanism has emerged a prominent area research. This paper delves mechanisms along with interrelationship. It specifically emphasizes core molecules within shared pathways governing neuroinflammation, including SIRT1, Nrf2, NF-κB, Cox-2, iNOS/NO·, how different immune cells structures contribute dysfunction through mechanisms. Researchers’ findings suggest that mutually promote each other may represent key factors progression disorders. A deeper comprehension common pathway between cellular holds promise for improving symptoms prognosis related

Depression is a major global health issue that urgently requires innovative and precise treatment options. In this context, saikosaponin has emerged as promising candidate, offering variety of therapeutic benefits may be effective in combating depression. This review delves into the multifaceted potential saikosaponins alleviating depressive symptoms. We summarized effects on structural functional neuroplasticity, elaborated regulatory mechanism modulating key factors affect such inflammation, hypothalamic-pituitary-adrenal (HPA) axis, oxidative stress, brain-gut axis. Moreover, paper highlights existing gaps current researches outlines directions for future studies. A detailed plan provided clinical application saikosaponins, advocating more targeted to speed up its transition from preclinical trials practice.

The incidence rate of depression, a mental disorder, is steadily increasing and has the potential to become major global disability factor. Given complex pathological mechanisms involved in use conventional antidepressants may lead severe complications due their side effects. Hence, there critical need explore development novel antidepressants. Ferroptosis, newly recognized form cell death, been found be closely linked onset depression. Several studies have indicated that certain active ingredients can ameliorate depression by modulating ferroptosis signaling pathway. Notably, traditional Chinese medicine (TCM) TCM prescriptions demonstrated promising antidepressant effects previous investigations owing unique advantages therapy. Building upon these findings, our objective was review recent relevant research provide new insights directions for application innovative strategies.
