Ulcerative colitis is a chronic intestinal disease linked to barrier damage, ferroptosis and dysbiosis. Coix lacryma-jobi natural food with food-medicine homology, whose seed-derived oil (Coix seed oil, CSO) has been shown anti-inflammatory activity in vitro. Here, the effects mechanisms of CSO on ulcerative (UC) vivo are systematically investigated. Firstly, UC mice was replicated by 3% DSS, assessed efficacy observing fecal occult blood, colon length, DAI score pathological histomorphological changes tissues. The barrier-protective were observed AB staining qRT-PCR. Secondly, biological targets obtained from TCMSP database Swiss Target Prediction database, downloaded FerrDb platform, UC-related GEO intersection above three taken obtain "CSO-UC-Ferroptosis" targets, which analysed GO KEGG enrichment, GSEA analysis, immune cell infiltration validation. Finally, core genes molecular docking potential active components CSO. In order further verify effect ferroptosis, GPX4 agonist RSL-3 used stimulate vivo, levels Iron, MDA SOD measured, immunohistochemistry detect tight junction proteins protein mice. Besides, evaluated intercellular junctions tissues each group under electron microscope. addition, 16sRNA sequencing performed contents observe flora improved physiological parameters, reduced inflammation response regulated restored gut microbiota balance Bioinformatics results showed that G6PD, ABCC1 at CSO, also demonstrated similar expression DSS-induced models vivo. Our findings demonstrate for first time ameliorated regulating mice, suggesting as promising candidate treatment warranting investigation.

Oxidative stress (OS) is a key factor in the generation of various pathophysiological conditions. Nuclear erythroid 2 (NF-E2)-related (Nrf2) major transcriptional regulator antioxidant reactions. Heme oxygenase-1 (HO-1), gene regulated by Nrf2, one most critical cytoprotective molecules. In recent years, Nrf2/HO-1 has received widespread attention as regulatory pathway for intracellular defense against oxidative stress. It considered potential target treatment inflammatory bowel disease (IBD). This review highlights mechanism action and therapeutic significance IBD complications (intestinal fibrosis colorectal cancer (CRC)), well phytochemicals targeting IBD. The results suggest that effects on mainly involve following aspects: (1) Controlling to reduce intestinal inflammation injury; (2) Regulation flora repair mucosal barrier; (3) Prevention ferroptosis epithelial cells. However, due complex role Nrf2/HO-1, more nuanced understanding exact mechanisms involved way forward future.

Ulcerative colitis (UC), a subtype of inflammatory bowel disease, manifests with symptoms such as abdominal pain, diarrhea, and mucopurulent, bloody stools. The pathogenesis UC is not fully understood. At present, the incidence has increased significantly around world. Conventional therapeutic arsenals are relatively limited, often poor efficacy many adverse effects. In contrast, traditional Chinese medicine (TCM) holds promise due to their notable effectiveness, reduced recurrence rates, minimal side recent years, significant progress been made in basic research on TCM for treatment. It found that inhibition ferroptosis through intervention can promote intestinal mucosal healing reverse UC. mechanism action involves multiple targets pathways. Aim review This summarizes experimental studies targeted regulation by its impact aiming provide theoretical basis prevention, treatment, further drug development Results Ferroptosis disrupts antioxidant mechanisms epithelial cells, damages mucosa, participates pathological process acts various pathways Nrf2 / HO-1 GSH/GPX4, blocking progression inhibiting damage thereby alleviating Conclusion diverse array single herbs, extracts herbal formulas facilitates selective innovative new methods targeting Although studying compound formulas, extracts, there still issues clinical designs, necessitating in-depth scientific exploration research.

Inflammatory bowel disease, particularly ulcerative colitis (UC), poses significant treatment challenges due to its chronic nature and potential for severe complications. This editorial reviews a recent network meta-analysis that evaluated the efficacy of highlighted superior outcomes achieved by combining each five traditional Chinese medicine (TCM) formulations with mesalazine adjuvant UC. Clinical included enhanced mucosal healing, improved quality life, reduced recurrence rates. Additionally, combination therapy resulted in lower incidence adverse reactions compared monotherapy. Despite these promising results, limitations such as variability study TCM dosage highlight need further high-quality, large-sample, multicenter randomized controlled trials. underscores enhancing UC management calls more rigorous research substantiate findings refine clinical guidelines.
