Quercetin is a natural polyphenolic flavonoid widely found in plants, fruits, and vegetables, has been reported to play pharmacological roles numerous pathogenic conditions. The anti-inflammatory effects of quercetin various inflammatory conditions diseases have well-documented. However, its regulatory role noncanonical inflammasome activation not yet demonstrated. This study investigated the caspase-11 inflammasome-activated responses macrophages mouse model acute lethal sepsis. protected J774A.1 from lipopolysaccharide (LPS)-induced cell death inflammasome-induced pyroptosis. It significantly decreased production mRNA expression pro-inflammatory cytokines, such as interleukin (IL)-1β, IL-18, IL-6, but tumor necrosis factor (TNF)-α, molecules, nitric oxide (NO) inducible NO synthase cells. Mechanistically, strongly suppressed autoproteolysis secretion proteolysis gasdermin D did inhibit direct binding LPS. In vivo, revealed that increased survival rate mice with sepsis serum levels cytokines without causing significant toxicity. conclusion, this highlights quercetin-mediated action through novel mechanism targets macrophages, suggesting promising agent medicine.

Inflammatory lung diseases such asthma, chronic obstructive pulmonary disease (COPD), acute respiratory distress syndrome (ARDS), injury, fibrosis arise from inflammation damage. While role inflammasomes in these well demonstrated, recent findings emphasize critical roles regulating various conditions. Particularly, new studies highlight their involvement diseases. The anti-platelet activity of the saponin fraction Korean Red Ginseng has been widely studied. consists panaxadiol (PDF) and panaxatriol (PTF); however, their is yet to be compared. Our study aimed investigate potency PDF PTF elucidate how well they retain via different administration routes. For ex vivo studies, Sprague-Dawley rats were orally administered 250 mg/kg for 7 consecutive days before blood collection cardiac puncture. Platelet aggregation was conducted after isolation washed platelets. in vitro platelets obtained from rats. Collagen adenosine diphosphate (ADP) used induce platelet aggregation. as an agonist assaying triphosphate release, thromboxane B2, serotonin, cyclic monophosphate, guanosine monophosphate (cGMP) release. When treated vivo, not only inhibited ADP collagen-induced aggregation, but also upregulated cGMP levels reduced adhesion fibronectin. Furthermore, it Akt phosphorylation induced by collagen treatment. Panaxadiol did exert any vitro, whereas exhibited potent activity, inhibiting ADP, collagen, thrombin-induced significantly elevated cGMP. showed that treatments levels, indicating possible metabolic conversions ginsenosides, which altered content ginsenosides capable preventing
