Abstract Menispermaceae species, as early‐diverging eudicots, can synthesize valuable benzylisoquinoline alkaloids (BIAs) like bisbenzylisoquinoline (bisBIAs) and sinomenines with a wide range of structural diversity. However, the evolutionary mechanisms responsible for their chemo‐diversity are not well understood. Here, chromosome‐level genome assembly Menispermum dauricum is presented demonstrated occurrence two whole duplication (WGD) events that shared by Ranunculales specific to , providing model understanding chromosomal evolution in eudicots. The biosynthetic pathway diverse BIAs M. reconstructed analyzing transcriptome metabolome. Additionally, five catalytic enzymes – one norcoclaurine synthase (NCS) four cytochrome P450 monooxygenases (CYP450s) from formation skeleton, hydroxylated modification, C‐O/C‐C phenol coupling BIAs. Notably, novel leaf‐specific MdCYP80G10 enzyme catalyzes C2′‐C4a ( S )‐reticuline into sinoacutine, enantiomer morphinan compounds, predictable stereospecificity discovered. Moreover, it found ‐specific CYP80 gene expansion, tissue‐specific expression, has driven BIA diversity compared other species. This study sheds light on WGD occurrences eudicots biosynthesis.

The challenge of discriminating closely related species persists, notably within clinical diagnostic laboratories for invasive aspergillosis (IA)-related and food contamination microorganisms with toxin-producing potential. We employed Analysis the whole-GEnome (AGE) to address challenges genus Aspergillus developed a rapid detection method. First, reliable whole genome data 77 were downloaded from database, through bioinformatic analysis, specific targets each identified. Subsequently, sequencing was validate these targets. Additionally, we an on-site method targeting target using editing system. Our results indicate that AGE has successfully achieved identification all IA-related ( fumigatus, niger, nidulans, flavus , terreus ) three well-known A. flavus, parasiticus oryzae section. Flavi have provided species-level-specific . Based on reference targets, substantiate efficacy distinguishing focal its species. Notably, amalgamation room-temperature amplification techniques demonstrates capacity accurate genomic DNA samples at concentration as low 0.1 ng/μl concise 30-min timeframe. Importantly, this methodology circumvents reliance large specialized instrumentation by presenting singular tube operational modality allowing visualized result assessment. These advancements aptly meet exigencies requirements specified species, facilitating prompt diagnosis quality monitoring. Moreover, based species-specific sequences, shows promising potential effective tool epidemiological research classification.

Corydalis yanhusuo W.T. Wang is a traditional herb. Benzylisoquinoline alkaloids (BIAs) are the main pharmacological active ingredients that play an important role in sedation, relieving pain, promoting blood circulation, and inhibiting cancer cells. However, there few studies on biosynthetic pathway of benzylisoquinoline , especially some specific components, such as tetrahydropalmatine. We carried out widely targeted metabolome transcriptomic analyses to construct identified candidate genes. In this study, 702 metabolites were detected, including 216 alkaloids. Protoberberine-type aporphine-type chemical components C . bulbs. Key genes for biosynthesis, 6-OMT CNMT NMCH BBE SOMT1 CFS SPS STOX MSH TNMT P6H successfully identified. There was no significant difference content expression level between two suborgans (mother-bulb son-bulb). The levels BIA expansion stage (MB-A SB-A) significantly higher than those maturity (MB-C SB-C), consistent with pattern gene regulation. Abstract In Traditional Chinese Medicine (TCM), the medicinal leech is vital for treatments to promote blood circulation and eliminate stasis. However, prevalence of counterfeit products in market undermines quality efficacy these remedies. DNA barcoding techniques, such as COI barcode, have been limited their application due amplification challenges. This study identified high variability 16 S rRNA gene within mitochondrial genome across five species, leading development a novel 219 bp mini-barcode. Compared with traditional our mini-barcode showed remarkable identification efficiency, classifying 142 out 147 samples from fresh processed materials. contrast, barcode could only successfully identify 79 samples. case seven batches decoction pieces, six, whereas recognized one. Additionally, effectively discerned species patent medicines when combined metabarcoding technology. These results confirm mini-barcode’s potential reliable tool rapidly precisely identifying TCM products.
