Parkinson's disease (PD) is a widespread neurodegenerative disorder that exerts broad variety of detrimental effects on people's health. Accumulating evidence suggests mitochondrial dysfunction, neuroinflammation, α-synuclein aggregation and autophagy dysfunction may all play role in the development PD. However, molecular mechanisms behind these pathophysiological processes remain unknown. Currently, research PD has focussed high mobility group box 1 (HMGB1), different laboratory approaches have shown promising outcomes to some level for blocking HMGB1. Given HMGB1 regulates participates modulates apoptosis, it hypothesised significance onset In current review, targeting multiple roles pathology was integrated, issues need future attention targeted therapeutic are mentioned.

Pulmonary hypertension (PH) is a progressive, pulmonary vascular disease with high morbidity and mortality. Unfortunately, the pathogenesis of PH complex remains unclear. Existing studies have suggested that inflammatory factors are key in PH. Interleukin-6 (IL-6) multifunctional cytokine plays crucial role regulation immune system. Current reveal IL-6 elevated serum patients it negatively correlated lung function those patients. Since one most important mediators inflammation PH, signaling mechanisms targeting may become therapeutic targets for this disease. In review, we detailed potential accelerating process specific pathways. We also summarized current drugs these pathways to treat hope study will provide more theoretical basis targeted treatment future.

Since the first 70 years of reporting cancer chemotherapy, malignant tumors have been second most common cause death in children and adults. Currently, commonly used anti-cancer methods include surgery, radiotherapy, immunotherapy. Although these treatment could alleviate cancer, they lead to different forms side effects no particularly significant effect on prolonging patients’ life span. Glycyrrhizic acid (GL), a native Chinese herbal extract, has wide range pharmacological effects, such as anti-cancer, anti-inflammatory, antioxidant, immune regulation. In this review, mechanisms GL are summarized various cancers. The inhibition chemotherapy-induced including hepatotoxicity, nephrotoxicity, genotoxicity, neurotoxicity pulmonary toxicity, is highlighted. Therefore, may be promising ideal drug for therapy.

For patients with epilepsy, antiseizure medication remains the primary treatment; however, it is ineffective in approximately 30% of cases. These experience progressive neuronal damage and poor outcomes. Therefore, there an urgent need for disease-modifying therapy (DMT) that targets pathogenesis epilepsy. Glycyrrhizin has shown potential as a DMT epilepsy due to its multiple diverse mechanisms. Previous studies suggest glycyrrhizin may regulate key processes involved pathogenesis, such neuroinflammation cell death, but effects on pyroptosis have not been reported. This study employed bioinformatics techniques identify molecular treatment then validated using kainic acid-induced status epilepticus mouse model. significantly prolonged seizure latency, reduced duration, alleviated Molecular experiments indicated through mediation high mobility group box 1 (HMGB1)/Toll-like receptor 4 (TLR4)/nuclear factor kappa-B (NF-κB) signaling pathway. exerts neuroprotective anticonvulsant by regulating via HMGB1/TLR4/NF-κB pathway, offering novel insights into

Brain microvascular endothelial cells are connected by tight junction (TJ) proteins and interacted adhesion molecules, which participate in the selective permeability of blood–brain barrier (BBB). The disruption BBB is associated with progression cerebral diseases. Pterostilbene a natural compound found blueberries grapes wide range biological activities, including anti-inflammatory, antioxidant, anti-diabetic effects. In this study, we investigated protective effects pterostilbene on LPS-stimulated mouse brain (bEnd.3) underlying mechanisms. results showed that effectively upregulated expressions such as zonula occludens (ZO)-1 claudin-5 downregulated expression molecules intercellular molecule (ICAM)-1 vascular cell (VCAM)-1, preventing damage under LPS stimulation. decreased LPS-triggered inducible nitric oxide synthase (iNOS) cyclooxygenase (COX)-2 well levels interleukin (IL)-6, tumor necrosis factor (TNF)-α (NO). Meanwhile, exerted an inhibitory effect nuclear kappa B (NF-κB) mitogen-activated protein kinase (MAPK) pathways bEnd.3 upon Additionally, exhibited antioxidant activating heme oxygenase 1 (HO-1). These findings indicated protected against lipopolysaccharide (LPS)-induced inflammation, oxidative stress (BBB) cells.
