Abstract
The
chemokine
receptor
CXCR3
is
traditionally
recognized
for
its
role
in
immune
cell
trafficking.
However,
emerging
evidence
suggests
that
functions
may
extend
beyond
the
system,
particularly
cancer,
where
roles
remain
to
be
elucidated.
In
this
study,
we
demonstrated
expression
correlates
with
glioblastoma
(GBM)
grading,
CXCR3-A
isoform
being
associated
poorer
patient
prognosis
compared
CXCR3-B.
Ablation
of
both
isoforms
significantly
impaired
GBM
proliferation,
migration,
and
tumor
growth
vitro
immunodeficient
mice.
To
elucidate
mechanistic
CXCR3,
conducted
transcriptomic
profiling
xenografts,
revealing
depletion
would
disrupt
mitochondrial
homeostasis.
This
was
further
supported
by
our
findings
localize
membrane,
inhibition
lead
depolarization
increased
reactive
oxygen
species
production.
Notably,
activation
phosphorylated-STAT3
rescued
viability
CXCR3-depleted
cells,
suggesting
modulate
function
through
a
STAT3-dependent
mechanism,
consistent
known
functional
STAT3
maintaining
redox
balance.
Furthermore,
treatment
selective
antagonist
AMG487
reduced
disrupted
vitro,
vivo,
patient-derived
stem
cells.
Our
reveal
as
previously
unrecognized
regulator
cancer
positioning
CXCR3-mitochondrial
signaling
axis
promising
therapeutic
target
GBM.
