Abstract The biofilm acts as a protective layer for Streptococcus suis ( S. ), contributing to the development of drug resistance and chronic infections. Autoinducer 2 (AI-2) quorum sensing represents primary regulatory pathway governing formation in . Consequently, targeting AI-2 inhibit promising strategy preventing managing infections caused by This study established small natural product library integrating commercial molecules with Chinese herbal medicine molecules. two products, salvianolic acid A (SAA) rhapontin (RH), which target via sensing, were identified. SAA RH synthesis through noncompetitive competitive binding S-ribosylhomocysteinase (LuxS). By inhibiting these compounds modulate expression adhesion genes extracellular polysaccharides (EPS), reducing ability ultimately formation. Using LC‒MS/MS, we further analysed impact on metabolic activity , revealing potential medicinal value compounds. Finally, efficacy against infection was validated Galleria mellonella larvae, confirming their significant anti-infection effects.
