Substance use disorder (SUD) is a chronic, relapsing disease with highly multifaceted pathology that includes (but not limited to) sensitivity to drug-associated cues, negative affect, and motivation maintain drug consumption. SUDs are prevalent, 35 million people meeting criteria for SUD. While addiction studied, most investigations of examine in isolation, rather than the more prevalent context comorbid substance histories. Indeed, 11.3% individuals diagnosed SUD have concurrent alcohol illicit disorders. Furthermore, having one increases susceptibility developing dependence on additional substances. For example, increased risk heroin twofold misusers, threefold cannabis users, 15-fold cocaine 40-fold prescription misusers. Given prevalence associated polysubstance current public health crises, examining these disorders through lens co-use essential translatability improved treatment efficacy. The escalating economic social costs continued rise has spurred interest preclinical models effectively model this phenomenon. Here, we review state field understanding behavioral neural circuitry common pairings alcohol, nicotine, cannabis, other addictive Moreover, outline key considerations when models, including challenges provide insights improve outcomes.

The role of the prefrontal cortex (PFC) takes center stage among unanswered questions in modern neuroscience. PFC has a Janus-faced nature: it enables sophisticated cognitive and social abilities that reach their maximum expression humans, yet underlies some devastating symptoms psychiatric disorders. Accordingly, appropriate development is crucial for many high-order dysregulation this process been linked to various neuropsychiatric diseases. Reviewing recent advances field, with primary focus on rodents we highlight why, despite differences across species, cross-species approach fruitful strategy understanding development. We briefly review developmental contribution molecules extensively discuss how electrical activity controls early maturation wiring areas, as well emergence refinement input-output circuitry involved processing. Finally, mechanisms dysfunction relevance

Despite dysfunctional vasoactive intestinal polypeptide-positive interneurons (VIP-INs) being linked to the emergence of neurodevelopmental disorders, temporal profile VIP-IN functional maturation and cortical network integration remains unclear. Postnatal development was traced with patch clamp experiments in somatosensory cortex Vip-IRES-cre x tdTomato mice. Age groups were chosen during barrel field formation, before after activation main sensory inputs, adult animals (postnatal days (P) P3-4, P8-10, P14-16, P30-36). Changes passive active membrane properties show a towards accelerated signal integrations. Excitatory inhibitory postsynaptic currents (EPSCs IPSCs) showed progressive into networks, likely via synaptogenesis: mEPSC frequency increased while mIPSC at P14-16. Only kinetics became accelerated, E/I ratio synaptic defined as charge transfer, remained constant throughout investigated developmental stages. Both the amygdala and bed nucleus of stria terminalis (BNST) have been implicated in maladaptive anxiety characteristics disorders. However, underlying circuit cellular mechanisms remained elusive. Here we show that mice with Erbb4 gene deficiency somatostatin-expressing (SOM+) neurons exhibit heightened as measured elevated plus maze test open field test, two assays commonly used to assess anxiety-related behaviors rodents. Using a combination electrophysiological, molecular, genetic, pharmacological techniques, demonstrate abnormal mutant is caused by enhanced excitatory synaptic inputs onto SOM+ central (CeA), resulting reduction inhibition downstream BNST. Notably, our results indicate an increase dynorphin signaling CeA mediates paradoxical BNST neurons, consequent activity both necessary for sufficient drive anxiety. Finally, associated dysfunctions increased CeA-BNST can be recapitulated stress wild-type mice. Together, unravel previously unknown processes extended cause anxiety.SIGNIFICANCE STATEMENT The has behaviors, but are unclear. found (CeA) controls through modulation terminalis, process mediated CeA.
