Cortisol and corticosterone (CORT) coordinate circadian events manage the stress response by differential activation of two complementary brain receptor systems, i.e., mineralocorticoid (MR) glucocorticoid (GR), which mediate rapid non-genomic slow genomic actions. Several recent discoveries are highlighted from molecular fine-tuning MR/GR balance FKBP5 to CORTs role in neural network regulation underlying adaptation emotional, cognitive, social domains behavior. Rett Syndrome, a neurodevelopmental disorder caused by loss-of-function mutations in the MECP2 gene, is characterized severe motor, cognitive and emotional impairments. Exposure to early stressful events increases susceptibility post-traumatic stress disorder (PTSD) in adulthood, which the hypothalamic-pituitary-adrenal (HPA) axis plays a crucial role. Studies have found that environmental enrichment (EE) mitigates detrimental outcomes of adversity. However, HPA-related mechanism remains unclear. In this study, we used single prolonged (SPS) paradigm explore long-term effects adolescent on behavior, HPA activity, as well expression levels glucocorticoid receptor (GR), mineralocorticoid (MR), corticotropin-releasing hormone 1 (CRF1R) and CRF2R hypothalamus hippocampus. Meanwhile, protective EE intervention were examined.
