Osteosarcoma, the most common primary bone cancer in children and adolescents, presents significant challenges, particularly metastasis recurrence, resulting poor survival rates. This study explores role of PANoptosis—a complex cell death mechanism involving pyroptosis, apoptosis, necroptosis—in osteosarcoma by identifying relevant long non-coding RNAs (lncRNAs) their prognostic significance. Bioinformatics analyses used RNA expression data from GEO TARGET databases to identify differentially expressed genes (DEGs) PANoptosis-related (PRGs). Co-expression analysis revealed lncRNAs linked PRGs, forming a risk model. Five PRGs two were significantly associated with prognosis, model showing high predictive accuracy (AUC 0.876, 0.787, 0.794 for 1, 3, 5 years). Notably, lncRNA AC133552.2 was downregulated tissues, correlating reduced immune infiltration. Silencing promoted proliferation migration, while overexpression inhibited tumor growth metastasis, confirmed xenograft models. emerges as potential biomarker therapeutic target, future research needed explore its molecular mechanisms clinical application.

Sorafenib, a first-line therapeutic option for advanced hepatocellular carcinoma (HCC), faces formidable challenge in the form of emerging resistance. Recently, oncogene DDX11 antisense RNA 1 (DDX11-AS1) has been implicated various cancers, including HCC. However, its role sorafenib resistance remains unknown. Our findings reveal that DDX11-AS1 is upregulated sorafenib-resistant HCC cells, contributing to their by suppressing ferroptosis. Further investigation elucidated mechanism which activates antioxidant Nrf2-Keap1 pathway. By interacting with Nrf2 and hindering association Keap1, enhances stability nuclear translocation Nrf2. In summary, our study unveils potent as an enhancer resistance, inhibiting sorafenib-induced ferroptosis through activation pathway These offer promising strategy overcome effectively treat

Background: The functional ramifications of internal N7-methylguanosine (m7G) modification on RNAs have recently come to light, yet its regulatory influence long noncoding (lncRNAs) during the inflammatory-carcinogenesis transformation process in hepatitis B virus (HBV)-mediated hepatocellular carcinoma (HCC) remains largely unexplored. Methods: Clinical surgical samples encompassing HBV-related HCC, comprising both HCC tissue (tumor group, HBV+) and corresponding adjacent liver (paracancerous HBV+), were collected for analysis. Additional normal tissues (normal HBV-) acquired from patients with hepatic hemangioma, serving as controls. Employing MeRIP-seq, differential m7G levels lncRNAs across these groups compared identify a subset exhibiting continuous dynamic changes modification. Subsequently, vitro validation was conducted. Results: A total 856 exhibited alterations when paracancerous tissue.
