The progression of tumors has been demonstrated to have a strong correlation with ferroptosis. Bis(4-hydroxy-3,5-dimethylphenyl) sulfone (TMBPS) shown effectively inhibit the proliferation hepatocellular carcinoma (HCC), but its underlying mechanism is not clear. In this study, ferrostatin-1 (Fer-1) was employed explore whether death HCC cells caused by TMBPS related intracellular lipid peroxides, Fe2+, malondialdehyde (MDA), GSH/GSSG, mitochondrial morphology, and potential were detected after treatment. target predicted molecular docking approach verified via quantitative real-time polymerase chain reaction (qRT-PCR), western blot, cellular heat transfer assay (CETSA). Our results revealed that Fer-1 reversed cell induced in cells. Treatment typical ferroptosis features, including increased levels MDA, along decreased GSSH/GSH ratio potential. These effects overexpressing SLC7A11. findings suggest triggered linked ferroptosis, potentially mediated through inhibition SLC7A11 expression.

Solute carrier (SLC) family transporters are crucial transmembrane proteins responsible for transporting various molecules, including amino acids, electrolytes, fatty and nucleotides. To date, more than fifty SLC transporter subfamilies have been identified, many of which linked to the progression hepatic steatosis fibrosis. These conditions often caused by factors such as non-alcoholic liver disease steatohepatitis, major contributors global burden. The activity members regulates transport substrates across biological membranes, playing key roles in lipid synthesis metabolism, mitochondrial function, ferroptosis. processes, turn, influence function hepatocytes, stellate cells, macrophages, thereby contributing development Additionally, some involved drug transport, acting critical regulators drug-induced steatosis. Beyond substrate certain also exhibit additional functions. Given pivotal role fibrosis, this review aimed summarize molecular mechanisms through these conditions.

ABSTRACT Epigenetic regulation in disease development has been witnessed within this decade. RNA methylation is the predominant form of epigenetic regulation, and most prevalent modification N6‐methyladenosine (m 6 A). Recently, emerged as a potential target for treatment. posttranscriptional gene expression that involved both physiological pathological processes. Evidence suggests m A significantly affects metabolism, its abnormal changes have observed variety diseases. Metabolic diseases are series caused by metabolic processes body, common include diabetes mellitus, obesity, nonalcoholic fatty liver disease, etc.; although pathogenesis these differs from each other to current understanding, recent studies suggested pivotal role modulating diseases, A‐based drug on agenda. This paper reviewed understanding hoping provide systematic information those area.

Endometriosis (EMS) is a chronic, estrogen-dependent inflammatory disease affecting 5–10% of women reproductive age, characterized by the growth endometrial tissue on outside uterus. The dysregulation iron metabolism leads to accumulation ions at lesion sites, resulting in oxidative stress and pro-inflammatory responses that promote progression EMS. mechanisms underlying ferroptosis EMS primarily involve accumulation, lipid peroxidation, loss glutathione peroxidase 4 activity. These confer resistance within ectopic tissues facilitate cell survival proliferation. Traditional Chinese medicine (TCM) has demonstrated therapeutic potential for modulating ferroptosis. Studies have shown TCM monomers may regulate transport proteins anti-oxidant defense mechanisms. formulas employ distinct treatment strategies depending stage EMS: early stages, they control growth, whereas later inhibit reduce inflammation order improve health slow progression. This study provides new perspective management summarizing role its pathological reviewing findings use regulating

Melanoma is the most aggressive type of skin cancer, with few therapeutic alternatives following metastasis development. In recent years, drug delivery-associated nanotechnology has shown promising targeted results diminished adverse effects compared to conventional treatments. This study aimed (1) examine plant-derived α-arbutin, a natural compound and (2) compare these findings bioactively developed liposomes containing α-arbutin utilizing B16-F10 murine melanoma cell line as model. Liposomes were obtained through reversed-phase evaporation by applying spray dryer assess their stability. The biologic assays measured cytotoxicity/antiproliferative (MTT, Neutral Red, dsDNA PicoGreen). addition, levels melanin purinergic enzymes also measured. production reactive oxygen species (ROS) nitric oxide (NO) was determined measure oxidative state. Treatment nano-liposome alpha-arbutin induced significant 68.4% cytotoxicity, similar positive control, in at 72 hr. Further, arbutin increased ROS nitrite formation hr highest concentration (100 300 µg/ml) Arbutin reduced all tested concentrations. lowered nucleotides (AMP, ADP, ATP) nucleoside (adenosine) cells. Evidence suggests that liposome can be considered an alternative immunosuppressive agent stimulated treatment.
