High stocking density (HSD) stress has detrimental effects on growth performance, intestinal barrier function, and microbiota in intensive animal production. Organic acids (OA) are widely used as feed additives for their ability to improve performance health poultry. However, whether dietary OA can ameliorate HSD stress-induced impaired broilers remains elusive. In this study, a total of 528 one-day-old male Arbor Acres were randomly allocated into 3 treatments with 12 replicates per treatment including 10 birds normal 17 HSD. The follows: 1) Normal + basal diet; 2) diets; 3) OA.HSD induce increased levels serum corticosterone, lipopolysaccharides, interleukin-1β, tumor necrosis factor-α, down-regulated mRNA expression ZO-1, resulting compromised (P < 0.05). Dietary could significantly reduce which accompanied by up-regulated interleukin-10, Moreover, down-regulate the TLR4 MyD88 inhibit NF-κB signaling pathway Additionally, decreased abundance Bacteroidetes disturbed balance microbial ecosystems, whereas restored disordered gut reducing competitive exploitative interactions communities Meanwhile, content acetic butyric acids, showed significant correlations inflammation indicators 0.05).Dietary ameliorated through restoring compositions induced elevating short-chain fatty acid production TLR4/NF-κB pathway. These findings demonstrated critical role mediating HSD-induced inflammatory responses, contributing exploring nutritional strategies alleviate animals.

Yinzhihuang granule (YZHG) has liver protective effect and can be used for clinical treatment of non-alcoholic fatty disease (NAFLD), but its material basis mechanism need to further clarified. This study aims reveal the YZHG treating NAFLD.Serum pharmacochemistry were employed identify components from YZHG. The potential targets against NAFLD predicted by system biology then preliminarily verified molecular docking. Furthermore, functional in mice was elucidated 16S rRNA sequencing untargeted metabolomics. From YZHG, 52 compounds identified, which 42 absorbed into blood. Network pharmacology docking showed that treats with multi-components multi-targets. improve levels blood lipids, enzymes, lipopolysaccharide (LPS), inflammatory factors mice. also significantly diversity richness intestinal flora regulate glycerophospholipid sphingolipid metabolism. Moreover, Western Blot experiment lipid metabolism enhance barrier function.YZHG may treat improving disruption enhancing barrier. This will reduce invasion LPS subsequently inflammation.

Abstract Background Oral infection with cysts is the main transmission route of Toxoplasma gondii ( T. ), which leads to lethal intestinal inflammation. It has been widely recognized that alters composition and metabolism gut microbiota, thereby affecting progression toxoplasmosis. However, potential mechanisms remain unclear. In our previous study, there was a decrease in severity toxoplasmosis after α-amylase (α-AMY) knocked out. Here, we established mouse models ME49 Δ α-amy cyst then took advantage 16S rRNA gene sequencing metabolomics analysis identify specific microbiota-related metabolites mitigate -induced inflammation analyzed underlying mechanism. Results There were significant differences between cyst- cyst-infected mice, transferring feces from mice infected into antibiotic-treated mitigated colitis caused by infection. showed relative abundances bacteria, such as Lactobacillus Bacteroides , Bifidobacterium [Prevotella] Paraprevotella Macellibacteroides enriched challenged cysts. Spearman correlation microbiota indicated some fatty acids, including azelaic acid, suberic alpha-linolenic acid (ALA), citramalic highly positively correlated identified bacterial genera. Both oral administration ALA fecal transplantation (FMT) decreased expression pro-inflammatory cytokines restrained MyD88/NF-κB pathway, ultimately improved host survival. Furthermore, treated reshaped colonization beneficial Enterobacteriaceae Proteobacteria Shigella Enterococcus . Conclusions The present findings demonstrate closely associated We provide first evidence can alleviate improving dysregulation suppressing production via pathway. Our study provides new insight medical application for treatment
