Circular RNAs (circRNAs) are a type of important non-coding that widely involve in the physiological and pathophysiological process. Recent research has established link between circHIPK3 malignant activity cancer cells. However, circHIPK3' role esophageal squamous cell carcinoma (ESCC) still needs more focus. To determine prognostic value patients with ESCC, expression was quantified 32 pairs ESCC using real-time polymerase chain reaction (RT-qPCR). In order to explore new prediction methods and key genes for gastric cancer. Firstly, we downloaded the 6 original sequencing data of cancer on Illumina HumanHT-12 platform from Array Expression Gene Omnibus, used bioinformatics identify 109 up-regulated 271 down-regulated genes. Further, performed univariate Cox regression analysis prognostic-related genes, then Lasso remove collinearity, finally multivariate analyze independent prognostic (MT1M, AKR1C2, HEYL, KLK11, EEF1A2, MMP7, THBS1, KRT17, RPESP, CMTM4, UGT2B17, CGNL1, TNFRSF17, REG1A). Based these, constructed a risk proportion signature, found that patients with high-risk have high degree malignancy. Subsequently, GSE15459 set verify signature. By calculating area under recipient operator characteristic curve 5-year survival rate, test verification are 0.739 0.681, respectively, suggesting signature has moderate ability. The nomogram is visualize sig-nature, calibration showed accuracy higher. Finally, verified expression prognosis hub gene, suggested KRT17 may be potential biomarkers.

Lactate blunts the anticancer immune response in breast cancer (BC). However, little is known about exact effect of lactate transporters such as monocarboxylate transporter 4 (MCT4) on immunotherapy. In this study, we investigated expression status and prognostic value MCT4 BC through large-scale transcriptome data. Our results showed that was overexpressed BC, particularly basal-like molecular subtype. Overexpression significantly correlated with high lesion grade poor prognosis. Enrichment analysis indicated MCT4-related genes were involved immune- metabolism-related bioprocesses, myeloid leukocyte activation, adaptive system, catabolic process. We also found lesions associated cell infiltration glycolytic rate-limiting enzymes like pyruvate kinase M2 (PKM2) hexokinases-3 (HK3). observations indicate may play a pivotal role maintenance tumor microenvironment (TIME) metabolic reprogramming. The glycolysis pathway (MCT4, PKM2, HK3) thus serve new targets to modulate TIME enhance immunotherapy efficiency.[Figure: see text].
