This study combines hyperspectral imaging technology with biochemical parameter analysis to facilitate the disease severity evaluation and early detection of lettuce downy mildew. The results reveal a significant negative correlation between index (DI) levels flavonoids (r = −0.523) anthocyanins −0.746), indicating role these secondary metabolites in enhancing plant resistance. Analysis data identified that spectral regions (410–503 nm, 510–615 630–690 nm) vegetation indices like PRI ARI2 were highly correlated DI, flavonoids, anthocyanins, providing potential indicators for assessment detection. Moreover, regression models developed using Partial Least Squares (PLS), Random Forest (RF), Convolutional Neural Network (CNN) algorithms demonstrated high accuracy reliability predicting highest R2 0.857, 0.910, 0.963, respectively. classification model PLS, RF, CNN successfully detected physiological changes within 24 h post-infection (highest 0.764), offering an effective tool key parameters PLS-DA model, PRI, also strong correlations DI. Maize Downy Mildew (MDM) is a devastating disease in the humid sub-tropical/tropical regions of Asia. In this study, prevalence MDM during rainy Kharif seasons south Karnataka state (India) ranged between 6.8% (2018) and 19.1% (2022). The research evaluated new fungicidal treatments assessed genetic tolerance maize lines to develop robust management strategies that enhance productivity stability. During 2021 2022, we conducted field trials evaluate effectiveness six different fungicides, both individually combination. most effective approach combined seed treatment with Metalaxyl (4%) Mancozeb (64%) WP, followed by foliar spray Azoxystrobin (18.2%) Difenoconazole (11.4%) SC. This reduced incidence 97.6% increased yield up 85.6 quintals per hectare, benefit-cost ratio 2.2. Additionally, screening 317 inbred 2019 identified 22 stable resistance over nine consecutive Rabi seasons, indicating their potential for sustained resistance. Liquid Chromatography-Mass Spectrometry (LC-MS) analysis revealed significant increases eighteen phenolic compounds fifteen flavonoid resistant genotypes. Specifically, genotypes exhibited elevated levels salicylic acid (4.2 9.2-fold), p-Coumaric (3.7 4.8-fold), o-Coumaric (4.5 7.4-fold), Caffeic (2.4 3.1-fold), Ferulic (2.3 2.8-fold). Flavonoid also increased, Naringenin ranging from 34.4 µg/g African Tall 130 MAI 224, Catechin 22.9 124.4 10, Epicatechin 1.3 8.2 10.
