AD is characterized by deposition amyloid beta (Aβ) plaque and tau-neurofibrillary tangles (TNTs) brain. The neuropathological changes are related plaques, neurofibrillary tangles, progression neuroinflammation, neuronal mitochondrial dysfunction, autophagy cholinergic synaptic dysfunction. Statins one main cornerstone drugs for management cardiovascular disorders regardless dyslipidemia status. Increasing use statins, mainly elderly groups primary secondary prevention diseases, may affect their cognitive functions. Extensive prolonged statins functions healthy subjects patients. Statins-induced impairments both patients health providers had been reported according post-marketing survey. This survey depends on sporadic cases, no measures were used. Evidence prospective observational studies gives robust conclusion regarding beneficial or detrimental effects Therefore, this study a narrative review aimed with evidences beneficial, detrimental, neutral AD.

Aim. To evaluate by meta-analysis of interventional studies the effect statin therapy on arterial wall inflammation. Background. Arterial exposure to low-density lipoprotein (LDL) cholesterol levels is responsible for initiation and progression atherosclerosis 18F-fluorodeoxyglucose Positron Emission Tomography-Computed Tomography (18F-FDG PET/CT) has been used detect inflammation monitor vascular anti-inflammatory effects lipid-lowering therapy. Despite a number statin-based exploring 18F-FDG uptake, these trials have produced inconsistent results. Methods. Trials with at least one treatment arm were searched in PubMed-Medline, SCOPUS, ISI Web Knowledge, Google Scholar databases. Target-to-background ratio (TBR), an indicator blood-corrected was as target variable activity. Evaluation biases, random-effects model generic inverse variance weighting, sensitivity analysis performed qualitative quantitative data assessment synthesis. Subgroup meta-regression analyses also performed. Results. Meta-analysis seven eligible studies, comprising 10 arms 287 subjects showed significant reduction TBR following (Weighted Mean Difference (WMD): −0.104, p = 0.002), which consistent both high-intensity (WMD: −0.132, 0.019) low-to-moderate intensity −0.069, 0.037). Statin dose/duration, plasma C-reactive protein level changes, baseline did not affect response statins. Conclusions. Statins effective reducing inflammation, assessed PET/CT imaging. Larger clinical should clarify whether either cholesterol-lowering or other pleiotropic mechanisms this effect.

Background: A wealth of evidence has shown that microRNAs (miRNAs) can modulate specific genes, increasing our knowledge on the fine-tuning regulation protein expression. miR-221 and miR-222 have been frequently identified as deregulated across different cancer types; however, their prognostic significance in remains controversial. In view these considerations, we performed an updated systematic review meta-analysis published data investigating effects miR-221/222 overall survival (OS) other secondary outcomes among patients. search PubMed, Web Knowledge, Cochrane Library databases was performed. Hazard ratios (HRs) 95% confidence intervals (95% CIs) were used to assess strength association. Results: Fifty studies, analyzing 6086 patients, included review. Twenty-five studies for 17 which assessed OS meta-analysis. High expression significantly predicted poor (HR: 1.48, CI: 1.14–1.93, p = 0.003 HR: 1.90, 1.43–2.54, < 0.001, respectively). Subgroup analysis revealed finding not robust one miR-222. Furthermore, high also associated with worse progression-free disease-free pooled recurrence-free survival. Conclusions: The demonstrated is prognosis whereas unclear. More work required fully elucidate role prognosis, particularly limitations existing results, including significant heterogeneity limited number some cancers.

Calcific aortic valve disease (CAVD) is highly prevalent with marked morbidity and mortality rates a lack of pharmaceutical treatment options because its mechanisms are unknown. Melatonin reported to exert atheroprotective effects. However, whether melatonin protects against calcification, whose pathogenesis shares many similarities that atherosclerosis, the underlying molecular remain In this study, we found intragastric administration for 24 weeks markedly ameliorated calcification in high cholesterol diet (HCD)-treated ApoE-/- mice, as evidenced by reduced thickness calcium deposition leaflets, improved echocardiographic parameters (decreased transvalvular peak jet velocity increased area), decreased osteogenic differentiation marker (Runx2, osteocalcin, osterix) expression valves. Consistent these vivo data, also confirmed suppression vitro hVICs. Mechanistically, level CircRIC3, procalcification circular RNA, which functions acting miR-204-5p sponge positively regulate gene dipeptidyl peptidase-4 (DPP4). Furthermore, CircRIC3 overexpression abolished inhibitory effects on hVIC differentiation. Taken together, our results suggest ameliorates via regulation CircRIC3/miR-204-5p/DPP4 signaling hVICs; therefore, medication might be considered novel strategy CAVD treatment.

Hypertension is a major harbinger of cardiovascular morbidity and mortality. It predisposes to higher rates myocardial infarction, chronic kidney failure, stroke, heart failure than most other risk factors. By 2025, the prevalence hypertension projected reach 1.5 billion people. The pathophysiology this disease multifaceted, as it involves nitric oxide endothelin dysregulation, reactive oxygen species, vascular smooth muscle proliferation, vessel wall calcification, among others. With advent new biomolecular techniques, various studies have elucidated gaping hole in etiology mechanisms hypertension. Indeed, epigenetics, DNA methylation, histone modification, microRNA-mediated translational silencing appear play crucial roles altering molecular phenotype into hypertensive profile. Here, we critically review experimentally determined associations between microRNA (miRNA) molecules pharmacotherapy. Particular attention given epigenetic underlying physiological responses antihypertensive drugs like candesartan, relevant clopidogrel, aspirin, statins Furthermore, how miRNA affects pharmaco-epigenetics especially highlighted.
