Caesarean section scar diverticulum (CSD) is a significant cause of infertility among women who have previously had section, primarily due to persistent inflammatory exudation associated with this condition. Even though abnormal bacterial composition identified as critical factor leading chronic inflammation, clinical data suggests that long-term cure often unattainable antibiotic treatment alone. In our study, we employed metagenomic analysis and mass spectrometry techniques investigate the fungal in CSD its interaction bacteria. We discovered local abnormalities can disrupt stability population entire microbial community by altering abundance via specific metabolites. For instance, Lachnellula suecica reduces several Lactobacillus spp ., such jensenii , diminishing production metabolites like Goyaglycoside A Janthitrem E . Concurrently, Clavispora lusitaniae Ophiocordyceps australis synergistically impact modulating metabolite abundance. Our findings underscore activity are key drivers dysbiosis CSD.

Gut microbes and their metabolites are actively involved in the development regulation of host immunity, which can influence disease susceptibility. Herein, we review most recent research advancements gut microbiota–immune axis. We discuss detail how microbiota is a tipping point for neonatal immune as indicated by newly uncovered phenomenon, such maternal imprinting, utero intestinal metabolome, weaning reaction. describe shapes both innate adaptive immunity with emphasis on short-chain fatty acids secondary bile acids. also comprehensively delineate disruption axis results immune-mediated diseases, gastrointestinal infections, inflammatory bowel cardiometabolic disorders (e.g., cardiovascular diabetes, hypertension), autoimmunity rheumatoid arthritis), hypersensitivity asthma allergies), psychological anxiety), cancer colorectal hepatic). further encompass role fecal transplantation, probiotics, prebiotics, dietary polyphenols reshaping therapeutic potential. Continuing, examine modulates therapies, including checkpoint inhibitors, JAK anti-TNF therapies. lastly mention current challenges metagenomics, germ-free models, recapitulation to achieve fundamental understanding regulates immunity. Altogether, this proposes improving immunotherapy efficacy from perspective microbiome-targeted interventions.

Abstract The presence of bacterial persisters is a key factor contributing to chronic infection. However, no effective treatment methods are currently available. Thus, platform developed, called reactive oxygen species (ROS) bomb, based on microenvironment‐adaptive hydrogel microspheres, oxidize the cell membranes in periprosthetic joint infection (PJI). Fenton reagent hydroxy iron oxide (FeOOH) and glucose oxidase (GOx)/calcium phosphate (CaP) acid‐responsive shell sequentially induced surface mesoporous polydopamine (PDA) nanoparticles by PDA‐mediated ion precipitation interfacial adhesion, followed coloaded with into microfluidic hyaluronic acid microspheres. Hydroxyl radicals explosively generated through GOx‐mediated oxidase, H 2 O production, its Fenton‐like reactions FeOOH, which also benefit from weakly acidic microenvironment around persisters, result destruction membrane, subsequent overflow cellular contents such as dsDNA, proteins, K + . bactericidal rates methicillin‐resistant Staphylococcus aureus epidermidis up 99.14% 98.96%, loads lesion location significantly decreased after ROS bombs treated, effectively alleviated inflammation bone resorption damage. This work provides new strategy toward clearance shows great application potential other infection‐related diseases.

The last decade of microbiome research has highlighted its fundamental role in systemic immune and metabolic homeostasis. plays a prominent during gestation into early life, when maternal lifestyle factors shape development the newborn. Breast milk further shapes gut colonization, supporting tolerance to commensal bacteria harmless antigens while preventing outgrowth pathogens. Environmental microbial that disrupt this process can dysregulate homeostasis, predisposing infants atopic disease childhood asthma. In health, low-biomass lung microbiome, together with inhaled environmental constituents, establishes immunological set point is necessary maintain pulmonary defense. However, perturbations physiological processes allow upper respiratory tract act as reservoir pathogenic bacteria, which colonize diseased cause severe inflammation. Studying these host-microbe interactions diseases holds great promise stratify patients for suitable treatment regimens biomarker discovery predict progression. Preclinical studies show microbes are constant flux cell division death, releasing by-products, vesicles system protect against diseases. next major advances may come from testing utilizing clinical benefit exploiting predictive power by employing multiomics analysis approaches.

Introduction A large proportion of patients with inflammatory bowel disease (IBD) experience IBD-related conditions outside the gastrointestinal tract, termed extraintestinal manifestations (EIMs) which further decreases quality life and, in extreme cases, can be threatening. The pathogenesis EIMs remains unknown, and although gut microbiota alterations are a well-known characteristic IBD, its relationship sparsely investigated. This study aimed to compare IBD without EIMs.

There is growing interest in the use of microbial-seeding interventions to mitigate impacts prenatal antibiotics, C-section, and lack breastfeeding on mother-child microbe sharing. However, relative importance maternal vaginal vs. fecal microbiota this process unclear. Analyzing 16S rRNA sequences from five US birth cohorts, we found that became more similar as pregnancy progressed, both niches influenced child's microbiota. The contribution increased when sampling occurred later gestation. As children aged 5 years, their increasingly resembled mother's compared Patterns sharing appeared differ by antibiotic use, mode (C-section vaginal), breastfeeding. Our findings enhance understanding niche-specific may inform interventions. Metagenomic studies are needed identify specific shared strains.
