Abstract “Through space” palladium/hydrogen shift is an efficient strategy to achieve selective functionalization of a specific remote C−H bond. Compared with relatively extensive exploited 1,4‐palladium migration process, the relevant 1,5‐Pd/H was far less investigated. We herein report novel pattern between vinyl and acyl group. Through pattern, rapid access 5‐membered‐dihydrobenzofuran indoline derivatives has been achieved. Further studies have unveiled unprecedented trifunctionalization (vinylation, alkynylation amination) phenyl ring through 1,5‐palladium relayed decarbonylative Catellani type reaction. A series mechanistic investigations DFT calculations provided insights into reaction pathway. Notably, it that in our case prefers stepwise mechanism involving Pd IV intermediate.

A palladium-catalyzed multicomponent reaction involving o-bromobenzaldehydes and two different isocyanides was developed to assemble series of isoindolinones with spiroindolenine or azepinoindole skeletons. This sequential insertion features mild conditions, a wide substrate scope, high efficiency. Preliminary mechanistic study indicated that the difference in steric hindrance between isocyanide components is crucial when regulating sequence, whereas ligand also played an important role during whole process.

Comprehensive Summary We developed a novel Pd‐catalyzed carbonylation of ortho ‐alkenyl iodobenzenes with CO, affording diverse array 3‐arylindenones in good to excellent yields (up 94% yield). This methodology exhibits broad substrate scope and functional group compatibility. The synthetic utility was demonstrated by gram‐scale reaction, product derivatizations, the preparation an intermediate for PPARγ agonist.

Abstract This review aims to collect and analyze recent results with respect the use of varied palladium preparations in synthesis five‐membered nitrogen heterocycles including condensed derivatives. Results have been selected focus on studies last three years. Furthermore, a common feature all methods treated here is that nitrogen‐containing ring formed via closing appropriate starting materials. Selected examples discussed will reveal plethora products from small monocycles multi‐ring systems can be successfully accessed. Major features are wide product ranges, high yields stereoselectivities often achieved under mild reaction conditions.

Most fungi generate secondary metabolites essential to their growth or used as chemical defense systems. They produce a large variety of small molecules, notably alkaloids containing an isoindolinone unit in the form bicyclic cichorine-type molecules more less substituted, up extended, heavily substituted tri- tetra-cyclic equipped with one two prenyl side chains. An extended family than 170 derivatives produced by mycelial endophytic has been identified and analyzed. These include many series such andicichorines, aspergillinines, caputmedusins, clitocybins, daldinans, emerimidines, emeriphenolicins, entonalactams, erinaceolactams, erinacerins, fendlerinines, marilines, meyeroguillines, stachartins, stachybotrins, stachybotrysams, sterenins, xylactams, others compounds endowed pharmacological properties described here. The also includes dimers, potent fibrinolytic compounds, those designated Stachybotrys microspora triprenyl phenol (SMTP) lead product SMTP-7 (orniplabin) currently developed for treatment stroke. repertoire isoindolinone-based natural products is (in contrast that vascular plants) comprises marked anti-inflammatory, antiviral, anticancer activities. Comprehensive Summary Herein, a [1,2]‐phospha‐Brook rearrangement‐initiated palladium‐catalyzed cyclization reaction for base‐controlled selective synthesis of 2 H ‐isoindole‐1‐carboxamide and ‐isoindole‐1‐carbonitrile derivatives has been described.
