Abstract Mitochondria, one of the most important organelles, represent a crucial subcellular target for fundamental research and biomedical applications. Despite significant advances in design DNA nanotechnologies variety bio‐applications, dearth strategies that enable mitochondria targeting molecular imaging therapy remains an outstanding challenge this field. In Minireview, we summarize recent progresses on emerging application nanotechnology mitochondria‐targeted tumor treatment. Abstract Despite the significant potential of RNA‐cleaving DNAzymes for gene regulation, their application is limited by low therapeutic efficacy and lack cell‐specific control. Here, a DNAzyme‐based nanohybrid designed ultrasound (US)‐controlled, enzyme‐activatable mRNA regulation presented, enabling tumor cell‐selective combination therapy. The constructed coordination‐directed self‐assembly an enzymatically‐triggerable DNAzyme (En‐Dz) natural sonosensitizer hemoglobin (Hb). Controlled US exposure induces reactive oxygen species generation from Hb units, which not only facilitates efficient endosomal escape En‐Dz, but also promotes translocation specific enzyme nucleus to cytoplasm, thereby enhancing efficacy. Notably, enzyme‐triggered, spatiotemporally‐controlled activation En‐Dz's catalytic activity results in enhanced cancer‐cell selectivity treatment. Furthermore, enzyme‐activated sonodynamic therapy significantly enhances anti‐tumor both vitro vivo. Abstract Over the last three decades, significant advancements have been made in development of biosensors and bioassays that use RNA‐cleaving DNAzymes (RCDs) as molecular recognition elements. While early examples RCDs were primarily responsive to metal ions, past decade has seen numerous reported for more clinically relevant targets such bacteria, cancer cells, small metabolites, protein biomarkers. 5 years several RCD‐based also evaluated using either spiked biological matrixes or patient samples, including blood, serum, saliva, nasal mucus, sputum, urine, faeces, which is a critical step toward regulatory approval commercialization sensors.
