Rapid and sensitive antigen detection using a lateral flow immunoassay (LFIA) is crucial for diagnosing infectious diseases due to its simplicity, speed, user-friendly features. However, it remains critical issue explore specific biorecognition elements powerful signal amplification. In this study, taking SARS-CoV-2 as proof of concept, peptide, WFLNDSELIML, binding the spike (S) was identified by nonamplified biopanning method, which exhibited high affinity target, with dissociation constant 9.29 ± 1.55 nM. Molecular docking analysis reveals that peptide binds N-terminal domain S antigen. Then, capture probe angiotensin-converting enzyme 2 probe, peptide-based (pLFIA) without any antibody developed, polydopamine nanosphere (PDA)@MnO2 nanocomposite excellent oxidase-like activity used colorimetric label, exhibiting dual-mode remarkable amplification natural melanin on-demand nanozyme catalytic enhancement. The PDA@MnO2-based pLFIA capable detecting limit 8.01 pg/mL, 18.7 times lower than conventional tagged gold nanoparticles. Additionally, as-proposed can detect up 150 transduction units/mL pseudoviruses spiked in saliva samples. Given outstanding analytical performance, proposed may offer reliable option rapid diagnosis SARS-CoV-2.

The global spread of monkeypox has become a worldwide public healthcare issue. Therefore, there is an urgent need for accurate and sensitive detection methods to effectively control its spreading. Herein, we screened by phage display two peptides M4 (sequence: DPCGERICSIAL) M6 SCSSFLCSLKVG) with good affinity specificity virus (MPXV) B21R protein. To simulate the state peptide in avoid spatial obstacles peptide, GGGSK was added at C terminus named as M4a. Molecular docking shows that M4a are bound different epitopes hydrogen bonds salt-bridge interactions, respectively. Then, selected capture probe, carbonized polymer dots (CPDs) fluorescent colorimetric double-signal peptide/antigen/signal peptide-displayed sandwich ELISA triggered horseradish peroxidase (HRP) through simple internal filtration effect (IFE) constructed.
