Abstract Proteins secreted from maternal, fetal, and placental tissue are vital for processes like development, immunotolerance, fetal growth, associated with pregnancy complications, necessitating predictive biomarkers. In this study, we introduce an acetonitrile-based precipitation coupled solid-phase extraction that addresses limitations of current low throughput blood-based biomarker discovery workflows. Our method is efficient cost-effective, identified 433 protein groups, showing specific associations enrichment in reproductive tissues such as the placenta, breast, endometrium. Significant alterations proteins related to hormonal regulation, immune modulation, development throughout gestation were observed. This approach offers comprehensive insights into circulating proteome, but also provides a scalable solution larger studies context complications. TOC

Successful human pregnancy needs several highly controlled steps to guarantee an oocyte’s fertilization, the embryo’s pre-implantation development, and its subsequent implantation into uterine wall. Comprehensive protein profiling in intestinal tissues provides detailed information about the pathogenesis of colorectal cancer (CRC). This study quantified expression levels 92 oncology-related proteins tumors, paired para-carcinoma tissues, and remote normal from a cohort 52 CRC patients utilizing Olink technology. The proteomic profile closely resembled that while distinctly differing tumors. Among 68 differentially expressed (DEPs) identified between tumor WISP-1, ESM-1, TFPI-2 showed most pronounced alterations exhibited relatively strong correlations. These markers also presented highest AUC values for distinguishing tissue types. Bioinformatic analysis DEPs revealed plasma membrane PI3K-AKT signaling pathway were among enriched GO terms KEGG pathways. Furthermore, although is typically recognized as suppressor, both enzyme linked immunosorbent assay (ELISA) analyses have demonstrated its significantly elevated tumors compared with tissues. To best our knowledge, this first to proteome using work offers valuable insights into potential biomarkers therapeutic targets CRC, complementing circulating proteins.

We conduct a nested case-control study where 126 deaths, 162 age-sex-matched survivors 152 HIV-negative controls are analyzed 92 protein Olink Organ Damage panel by proximity extension assays (PEA). Using LASSO regression, logistic ROC analysis, twelve proteins significantly associated death, which six (SIRT5, PPM1B, PSMA1, GALNT10, VEGFC, PTN) specifically two (RCOR1, SERPINA9) four (CA12, CA14, RARRES1, EDIL3) both. Seasonal allergic rhinitis (SAR) is a prevalent inflammatory condition, yet its molecular mechanisms and reliable biomarkers remain incompletely understood. This study aimed to identify key inflammation-related proteins pathways associated with SAR by investigating seasonal proteomic profile variations their correlations symptoms. Serum samples were collected from nineteen patients during both allergy (in-season, IS) non-allergy (out-of-season, OS) periods. Differentially expressed (DEPs) identified using the Olink Target 96 Inflammation panel, which further analyzed through Gene Ontology (GO) Kyoto Encyclopedia of Genes Genomes (KEGG) pathway enrichment analyses. Spearman correlation analysis was conducted explore associations between DEPs symptoms, including sneezing, rhinorrhea, nasal blockage, itchy nose, eye. A total 36 identified, all significantly upregulated in season. Notable such as glial cell line-derived neurotrophic factor (GDNF), interleukin-18 receptor 1 (IL-18R1), interleukin-15 alpha (IL-15RA) showed strong Sneezing IL-2 beta (IL-2RB) (r = 0.415, p 0.013), rhinorrhea FMS-related tyrosine kinase 3 ligand (Flt3L) 0.455, 0.004), blockage osteoprotegerin (OPG) 0.493, 0.002). GO revealed enrichments Ras signaling small GTPase pathways, while KEGG highlighted immune-related PI3K-Akt cytokine-cytokine interactions. that vary seasonally SAR, offering insights into potential therapeutic targets for management. Proteomics has emerged as a transformative tool in biomedical research, enabling comprehensive characterization of protein profiles complex biological systems. In preeclampsia (PE) quantitative proteomic analyses plasma and serum have revealed critical insights into disease mechanisms potential biomarkers. Through systematic review 17 studies (2009–2024), we identified 561 differentially expressed plasma/serum proteins (p < 0.05) PE patients versus healthy controls, with 122 consistently replicated across ≥2 independent studies. Stratified analysis by clinical subtype (early-vs. late-onset PE) demonstrated both concordant divergent expression patterns, reflecting heterogeneity pathophysiology, methodological variations (e.g., sample processing, platforms), differences between discovery-phase targeted validation The trimester-specific biomarker panels proposed here offer framework for future large-scale, multicenter validation. By integrating advanced technologies standardized preanalytical analytical protocols, these findings advance opportunities early prediction (first-trimester signatures); mechanistic insight (complement system involvement); personalized management (subtype-specific therapeutic targets). This work underscores the proteomics to reshape from molecular discovery translation, ultimately improving outcomes this leading cause maternal perinatal morbidity.
