A highly oxygenated γ-pyrone with an unprecedented carbon skeleton, peunipyrone (1), was isolated from the sponge-derived fungus Aspergillus peuniceus. Compound 1 features a unique fused 6/6/6/6/6 pentacyclic ring system. The chemical structure of compound elucidated by comprehensive spectroscopic techniques and single-crystal X-ray diffraction. plausible biosynthetic pathway proposed. promoted leukemic cell apoptosis increasing p53 expression.

ABSTRACT Four new polyketides including one indanone ( 1 ), isobenzofuranone 2 and two naphthoquinones 3 4 ) were isolated from the deep‐sea‐derived fungus Pseudogymnoascus hyalinus , along with 20 known compounds 5 – 24 ). The structures of established by comprehensive analyses their 1D 2D NMR, HRESIMS, as well TD‐DFT‐ECD. Compounds 6 10 displayed potent inhibitory effects against RSL3 induced ferroptosis EC 50 values 4.74, 1.55, 3.72 µM, respectively.

Comprehensive Summary A chemical investigation of the deep‐sea‐derived fungus Penicillium allii‐sativi MCCC 3A00580 resulted in discovery four new meroterpenoids ( 1 — 4 ) and one related known co‐metabolite 5 ). These showcase unique carbon skeletons featuring a common drimane sesquiterpene part with highly diverse polyketide units. Particularly, compound incorporates salicylic acid moiety while 2 possesses rare peroxide bridge part. The structures compounds were assigned by extensive spectroscopic analysis, quantum calculations, biogenetic considerations. Notably, 3 significantly blocked mTOR signaling pathway, resulting arrest cell cycle at G0‐G1 phase triggering mitochondrial apoptosis Hela cells. While previously reported macrophorin (MPA) effectively triggered death MDA‐MB‐231 cancer cells activating pathways involving receptors, mitochondria, mTOR, TNF.
