Infections are regarded as the most severe complication associated with human health, which urgent to be solved. Stimuli-responsive materials appealing therapeutic platforms for antibacterial treatments, provide great potential accurate theranostics. In this review, advantages, response mechanisms, and key design principles of stimuli-responsive highlighted. The biomedical applications, current challenges, future directions also discussed. First, categories comprehensively itemized based on different sources stimuli, including external physical environmental stimuli (e.g., temperature, light, electricity, salt, etc.) bacterial metabolites acid, enzyme, redox, etc.). Second, structural characteristics, principles, applications responsive discussed, underlying interrelationships revealed. molecular structures closely related stimuli. Nitric oxide (NO), a gaseous transmitter extensively present in the human body, regulates vascular relaxation, immune response, inflammation, neurotransmission, and other crucial functions. Nitrite donors have been used clinically to treat angina, heart failure, pulmonary hypertension, erectile dysfunction. Based on NO's vast biological functions, it further can tumors, bacteria/biofilms infections, wound healing, eye diseases, osteoporosis. However, delivering NO is challenging due uncontrolled blood circulation release half-life of under five seconds. With advanced biotechnology development nanomedicine, packaged with multifunctional nanocarriers by physically embedding or chemically conjugating reported show improved therapeutic efficacy reduced side effects. Herein, we review discuss recent applications nanomedicines, their mechanisms, challenges nanomedicines for future scientific studies clinical applications. Abstract Reactive nitrogen species (RNS) generated via the reaction of nitric oxide (NO) with reactive oxygen (ROS) are more lethal than ROS, and thus RNS‐mediated therapy has great potential in cancer treatment, yet it is still largely unexploited. Herein, a novel, injectable NO‐releasing hydrogel (NO‐Gel) composed α‐(nitrate ester) acetic acid‐modified amphiphilic copolymers developed. To further convert released NO to RNS, glutathione (GSH)‐sensitive CuCys nanoparticles (NPs) β‐lapachone (Lapa) co‐loaded into NO‐Gel. This system possesses temperature‐induced sol‐gel transition can continuously liberate Lapa, NPs, vitro for up 3 weeks. The sustained supply Lapa efficiently boost hydrogen peroxide (H 2 O ) concentration cells, intracellular GSH induce rapid release reduction NPs. With elevating H levels producing highly Cu(I), Cu(I)‐catalyzed Fenton‐like dramatically enhanced, resulting generation abundant hydroxyl radicals (·OH), subsequent cascade reactions among ·OH, 2, cause RNS pool. After single peritumoral injection system, ROS plus substantial depletion significantly suppress tumor growth.

Abstract Compared to conventional photothermal therapy (PTT) which requires hyperthermia higher than 50 °C, mild‐temperature PTT is a more promising antitumor strategy with much lower phototoxicity neighboring normal tissues. However, the therapeutic efficacy of always restricted by thermoresistance cancer cells. To address this issue, supramolecular drug nanocarrier fabricated co‐deliver nitric oxide (NO) and agent DCTBT NIR‐II aggregation‐induced emission (AIE) characteristic for PTT. NO can be effectively released from nanocarriers in intracellular reductive environment capable simultaneously producing reactive oxygen species (ROS) upon 808 nm laser irradiation. The generated ROS further react produce peroxynitrite (ONOOˉ) bearing strong oxidization nitration capability. ONOOˉ inhibit expression heat shock proteins (HSP) reduce cells, necessary achieve excellent DCTBT‐based at mild temperature (<50 °C). performance ONOOˉ‐potentiated validated on subcutaneous orthotopic hepatocellular carcinoma (HCC) models. This research puts forward an innovative overcome PTT, provides new inspirations explore ONOOˉ‐sensitized tumor strategies.

Abstract Cancer cells typically display redox imbalance compared with normal due to increased metabolic rate, accumulated mitochondrial dysfunction, elevated cell signaling, and accelerated peroxisomal activities. This may regulate gene expression, alter protein stability, modulate existing cellular programs, resulting in inefficient treatment modalities. Therapeutic strategies targeting intra‐ or extracellular states of cancer at varying state progression trigger programmed death if exceeded a certain threshold, enabling therapeutic selectivity overcoming resistance radiotherapy chemotherapy. Nanotechnology provides new opportunities for modulating their excellent designability high reactivity. Various nanomaterials are widely researched enhance highly reactive substances (free radicals) production, disrupt the endogenous antioxidant defense systems, both. Here, physiological features described challenges illustrated. Then, that classified elaborated upon based on ability target regulations. Finally, future perspectives this field proposed. It is hoped review guidance design nanomaterials‐based approaches involving therapy, especially cancers resistant chemotherapy, etc.
