Abstract Molecular phototheranostic dyes are of eminent interest for oncological diagnosis and imaging‐guided phototherapy. However, it remains challenging to develop photosensitizers (PSs) that simultaneously integrate high‐contrast photoacoustic imaging efficient therapeutic capabilities. In this work, a supramolecular strategy is employed construct molecular pair agent via the direct self‐assembly two cyanines, C5TNa (anionic) Cy‐Et (cationic). The Coulombic interactions between facilitate formation complementary cyanine (C5T‐ET) creation CT‐J‐type aggregates in water. This results completely quenched fluorescence significantly enhances nonradiative deactivation (≈22 ps), leading 3.3‐fold increase photothermal conversion efficiency 7.1‐fold enhancement response compared indocyanine green (ICG). As result, J‐type aggregate demonstrates high capability remarkable antitumor efficacy vivo, highlighting its potential clinical applications. work provides strong experimental evidence superior performance enhancing photosensitization response. It believed will propel advancement controllable dye J‐aggregates contribute practical implementation phototherapy vivo.

However, developing straightforward strategies to activate stress-sensitive membrane channels situ poses significant challenges. In this study, we present a chemiluminescence resonance energy transfer (CRET) system based on conjugated oligomer, oligo(p-phenylenevinylene)-imidazolium (OPV-Im), designed the activation transient receptor potential melastatin 2 (TRPM2) calcium by superoxide anion (O2⋅-) without requiring external light sources. OPV-Im oligomer targeted cell efficiently, leading TRPM2 CRET process subsequent intracellular overload. This cascade resulted mitochondrial damage inhibition autophagy, ultimately inducing apoptosis. Additionally, strategy could be applied selective killing tumor that overexpress ion inhibiting growth three-dimensional (3D) spheroids. Our offers novel approach regulating channel activity compared optogenetics photodynamic therapy.
