The coronavirus disease 2019 pandemic, caused by severe acute respiratory syndrome 2 (SARS-CoV-2) coronavirus, is a global health issue with unprecedented challenges for public health. SARS-CoV-2 primarily infects cells of the tract via spike glycoprotein binding to angiotensin-converting enzyme (ACE2). Circadian rhythms coordinate an organism's response its environment and can regulate host susceptibility virus infection. We demonstrate that silencing circadian regulator Bmal1 or treating lung epithelial REV-ERB agonist SR9009 reduces ACE2 expression inhibits entry replication. Importantly, infected limits replication secretion infectious particles, showing post-entry steps in viral life cycle are influenced system. Transcriptome analysis revealed induced interferon-stimulated gene transcripts Calu-3 cells, providing mechanism pathway limit Our study highlights alternative approaches understand improve therapeutic targeting SARS-CoV-2.

Abstract The circadian system is responsible for internal functions and regulation of the organism according to environmental cues (zeitgebers). Circadian rhythm dysregulation or chronodisruption has been associated with several diseases, from mental autoimmune life quality change. Following this, some therapies have developed correct misalignments, such as light therapy chronobiotics. In this manuscript, we describe circadian‐related diseases so far investigated, studies reporting relevant data on topic, evidencing relationship, are included. Despite actual limitations in published work, there clear evidence correlation between disease origin/development, and, way, clock‐related emerge great progress clinical field. Future improvements interventions can lead development successful chronotherapy strategies, deeply contributing enhanced therapeutic outcomes.

The complement component C3 is a fundamental plasma protein for host defense, produced largely by the liver. However, recent work has demonstrated critical importance of tissue-specific expression in cell survival. Here, we analyzed effects local versus peripheral sources model acute bacterial pneumonia induced Pseudomonas aeruginosa . Whereas mice with global deficiency had severe pneumonia-induced lung injury, those deficient only liver-derived remained protected, comparable to wild-type mice. Human transcriptome analysis showed that secretory epithelial cells, such as club express high levels mRNA. Mice tamoxifen-induced gene ablation from cells worse pulmonary injury compared similarly treated controls, despite maintaining normal circulating levels. Last, both mouse and cultured primary human airway stress-induced death associated parallels seen Factor B rather than C3a receptor deficiency. Moreover, C3-mediated reduction requires alternative pathway B. Thus, our findings suggest reliant on locally derived protects mucosal barrier.

Abstract Patients admitted to the intensive care unit (ICU) are in need of continuous organ replacement strategies and specialized care, for example because neurological dysfunction, cardio‐pulmonary instability, liver or kidney failure, trauma, hemorrhagic septic shock even preterm birth. The 24‐h nursing interventions provided critically ill patients significantly limit resting and/or recovery phases. Consecutively, patient's endogenous circadian rhythms misaligned disrupted, which turn may interfere with their critical condition. A more thorough understanding complex interactions effectors tissue‐specific molecular clocks could therefore serve as potential means enhancing personalized treatment patients, conceivably restoring network thus accelerating physical neurocognitive recovery.
