This paper describes the design and synthesis of a photostable, far-red to near-infrared (NIR) platform for optical voltage sensing. We developed new, sulfonated silicon rhodamine fluorophore integrated it with phenylenevinylene molecular wire create Berkeley Red Sensor Transmembrane potential, or BeRST 1 ("burst"). is first member class NIR sensitive dyes that make use photoinduced electron transfer (PeT) trigger interrogation membrane voltage. show displays bright, membrane-localized fluorescence in living cells, high photostability, excellent sensitivity neurons. Depolarization plasma results rapid increases (24% ΔF/F per 100 mV). can be used conjunction fluorescent stains organelles, Ca(2+) indicators, voltage-sensitive proteins. In addition, red-shifted spectral profile 1, relative commonly employed optogenetic actuators like ChannelRhodopsin2 (ChR2), which require blue light, enables electrophysiology The speed, sensitivity, photostability long-wavelength profiles useful noninvasive, dissection neuronal activity.
