The highly immunosuppressive tumor microenvironment (TME) restricts the efficient activation of immune responses. To restore surveillance system for robust activation, vast efforts are devoted to normalizing TME. Here, a manganese-doped layered double hydroxide (Mn-LDH) is developed potent anti-tumor immunity by reversing Mn-LDH synthesized via one-step hydrothermal method. Sonodynamic therapy (SDT), which is non-invasive and controllable has the potential to treat triple-negative breast cancer (TNBC). However, hypoxia immunosuppressive tumor microenvironment (TME) often block production of reactive oxygen species induction SDT-activated immunogenic cell death, thus limiting activation adaptive immune responses. To alleviate these challenges, we proposed development a multifunctional biomimetic nanoplatform (mTSeIR), was designed with diselenide-conjugated sonosensitizers tirapazamine (TPZ), encapsulated within M1 macrophage membrane. This utilized hypoxia-induced chemotherapy improve efficacy SDT, further enhance immunotherapy by activating innate immunity remodeling TME. Firstly, prodrug TPZ activated due increased consumption associated SDT. Subsequently, mTSeIR enhanced repolarization M2 macrophages phenotype. The diselenide component in effectively natural killer cell-mediated antitumor response. Ultimately, vivo studies indicated that mTSeIR+US good biosafety achieved over 98% inhibition immunotherapy. research presents an efficient approach addressed limitations SDT achieves simultaneous both immunotherapy, resulting significant anti-metastatic TNBC.

Abstract Background In recent years, immunotherapy has made significant progress. However, the understanding of heterogeneity and function T cells, particularly CD8 + in cervical cancer (CESC) microenvironment remains insufficient. We aim to characterize heterogeneity, developmental trajectory, regulatory network, intercellular communication cells squamous cell carcinoma construct a prognostic risk model based on transcriptomic characteristics cells. Methods integrated single-cell RNA sequencing data from CESC tumor samples with bulk transcriptome TCGA GEO databases. identified subsets microenvironment, revealing interactions between other types through analysis. Pseudotime trajectory analysis revealed dynamic transcriptional regulation during differentiation functional acquisition processes. constructed network for identifying key transcription factors. Based cell-related genes, comprising eight core genes was developed validated using machine learning. Results four distinct subsets, namely progenitor, intermediate, proliferative, terminally differentiated, each exhibiting unique properties. interact macrophages different ligand-receptor networks, including CCL-CCR signaling pathway costimulatory molecules. Sorafenib as potential immunotherapeutic drug screening. Experimental validation demonstrated that sorafenib enhances cytotoxicity by increasing secretion IFN-γ TNF-α, thereby significantly inhibiting invasiveness survival Conclusions Our study provides valuable insights into diversity CESC. demonstrate signature may serve tool stratification patients Additionally, our finding suggests could be promising therapeutic candidate improving antitumor immunity this patient population.

Alcohol-associated liver disease (ALD) is a growing global health concern and its prevalence severity are increasing steadily. While bacterial endotoxin translocation into the portal circulation well-established key factor, recent evidence highlights critical role of sterile inflammation, triggered by diverse stimuli, in alcohol-induced injury. This review provides comprehensive analysis complex interactions within hepatic microenvironment ALD. It examines contributions both parenchymal cells, like hepatocytes, non-parenchymal such as stellate Kupffer neutrophils, sinusoidal endothelial driving progression disease. Additionally, we explored involvement mediators, including cytokines, chemokines inflammasomes, which regulate inflammatory responses promote injury fibrosis. A particular focus has been placed on extracellular vesicles (EVs) essential mediators intercellular communication beyond liver. These facilitate transfer signalling molecules, microRNAs proteins, modulate immune responses, fibrogenesis lipid metabolism, thereby influencing progression. Moreover, underscore importance organ-to-organ crosstalk, particularly gut-liver axis, where dysbiosis increased intestinal permeability lead to microbial translocation, exacerbating inflammation. The adipose-liver axis also highlighted, impact adipokines free fatty acids from adipose tissue steatosis inflammation context alcohol consumption.
