Abstract Suicide attempt (SA) risk is elevated in individuals with bipolar disorder (BD), and DNA methylation patterns may serve as possible biomarkers of SA. We conducted epigenome-wide association studies (EWAS) blood associated BD was measured at > 700,000 positions a discovery cohort n = 84 adults history SA (BD/SA), 79 without (BD/non-SA), 76 non-psychiatric controls (CON). EWAS revealed six differentially methylated (DMPs) seven regions (DMRs) between BD/SA BD/non-SA, multiple immune-related genes implicated. There were no significant differences when BD/non-SA each compared to CON, suggested that epigenetics differentiating from do not differentiate CON. Weighted gene co-methylation network analysis trait enrichment the vs. contrast further corroborated immune system involvement, while ontology implicated calcium signalling. In an independent replication 48 47 fold-changes cohort’s sites showed moderate correlation across cohorts agreement on direction. both cohorts, classification accuracy for among highest CpG well information clinical interviews combined, AUC 88.8% (CI 83.8-93.8%) 82.1% 73.6-90.5%) combined epigenetic-clinical predictor respectively. Abstract Objective Associations have been seen between suicidal behavior and differential DNA methylation of certain genes, with one study showing significant hypomethylation ARHGEF38 in postmortem brain samples from individuals bipolar disorder who died by suicide. Our objective was to explore a history suicide attempt. Method With pyrosequencing, we looked at the previously identified region interest ARHGEF38. We investigated levels 3 CpG sites 47 attempt, without non-bipolar controls. Results None measured had an association groups, although there were distinct clusters each group. Applying genotypes SNPs found interest, rs2121558 rs1447093, these showed stepwise site, regardless phenotype. Conclusions In this relatively small sample size study, not associated failing replicate findings related outcome, death. However, did provide evidence SNP interplay region. This highlights potential relevance considering genetics when interrogating epigenetic mechanisms. Bipolar disorder (BD) is a chronic and severe psychiatric associated with significant medical morbidity reduced life expectancy. In this study, we assessed accelerated epigenetic aging in individuals BD using various DNA methylation (DNAm)-based markers. For purpose, used five clocks (Horvath, Hannum, EN, PhenoAge, GrimAge) DNAm-based telomere length clock (DNAmTL). DNAm profiles were obtained Infinium MethylationEPIC Arrays from whole-blood samples of 184 BD. We also estimated blood cell counts based on levels for adjustment. Abstract Individuals with bipolar disorder are at increased risk for suicide, and this can be influenced by a range of biological, clinical, environmental factors. Biological components associated suicide include DNA modifications that lead to changes in gene expression. Common genetic variation methylation some the most frequent types findings an suicidal behavior. Importantly, interplay between predisposition patterns is becoming more prevalent studies. We hypothesized specific loci already genetically would altered individuals history attempt. To test hypothesis, we searched literature identify common variants (N=34) previously thoughts behaviors disorder. then created customized sequencing panel covered our chosen genomic loci. profiled from blood samples collected participants behavior (N=55) without (N=51). identified seven differentially methylated CpG sites five regions two groups. Additionally, found MIF CACNA1C were lethality or number attempts. Finally, three meQTLs SIRT1 , IMPA2 INPP1 . This study illustrates attempts known harbor suicide-related variants.

Telomere length (TL), mitochondrial DNA copy number (mtDNAcn), and methylation age (DNAmAge) are common aging biomarkers. However, research on the associations between these three markers at birth subsequent metabolic status was limited. This study aimed to evaluate association TL, mtDNAcn, DNAmAge in newborns variation hormones of children 3 years old. involved 895 mother-child pairs from a cohort China, with TL mtDNAcn measured using quantitative real-time PCR, (DNAm) assessed Infinium MethylationEPIC Beadchip, DNAm determined Horvath's epigenetic clock. Insulin leptin levels were via electrochemiluminescence assay. Multivariable adjusted linear regression restricted cubic spline (RCS) analysis utilized examine hormones. The indicated percentage change metabolism for per doubling biomarkers alterations found significant insulin (adjusted percent (95% CI), - 13.22 (- 23.21 1.94)), 15.32 (1.32 31.24)), 14.13 21.59 5.95)). RCS revealed non-linear (Ln transformed) (P = 0.024 nonlinearity), as well 0.043 nonlinearity). Specifically, insulin, positive observed when less than 0.05, which transitioned an inverse greater 0.05. Regarding leptin, there sharp decline 1.35, followed by plateau 1.35 0.67 then further 0.67. Abstract Background Bipolar disorder is a debilitating mood associated with high risk of suicide and characterized by immune dysregulation. In this study, we used multi-faceted approach to better distinguish the pattern dysregulation profiles in individuals BD. Methods We analyzed peripheral blood mononuclear cells (bipolar N=39, control N=30), serum cytokines N=86, N=58), whole RNA N=25, N=25), DNA N=104, N=66) identify immune-related differences participants diagnosed bipolar compared controls. Results Flow cytometry revealed higher proportion monocytes together lower T helper cells. Additionally, levels 18 were significantly elevated, while two reduced disorder. Most altered proinflammatory. Forty-nine genes differentially expressed our cohort further analyses uncovered several pathways these individuals. Genetic analysis indicated variants inflammatory bowel disease also influences risk. Discussion Our findings indicate significant component pathophysiology genetic overlap disease.
