Circular RNAs (circRNAs), covalently closed noncoding RNAs, are widely expressed in eukaryotes and viruses. They can function by regulating target gene expression, linear RNA transcription protein generation. The phosphoinositide 3-kinase (PI3K)/AKT signaling pathway plays key roles many biological cellular processes, such as cell proliferation, growth, invasion, migration, angiogenesis. It also a pivotal role cancer progression. Emerging data suggest that the circRNA/PI3K/AKT axis modulates expression of cancer-associated genes thus regulates tumor Aberrant regulation circRNAs is significantly associated with clinicopathological characteristics an important functions. In this review, we summarized functions PI3K-AKT-related vitro vivo assessed their associations characteristics. We further discussed diagnosis, prognostication, treatment cancers.

Background: Accumulating evidence indicates that non-coding RNAs (ncRNA), including long (lncRNAs) and circular (circRNAs), can function as competitive endogenous (ceRNAs) by binding to microRNAs (miRNAs) regulating host gene expression at the transcriptional or post-transcriptional level.Dysregulation in ceRNA network regulation has been implicated occurrence development of cancer. However, lncRNA/circRNA-miRNA-mRNA regulatory is still lacking nasopharyngeal carcinoma (NPC).Methods: Differentially expressed genes (DEGs) were obtained from our previous sequencing data Gene Expression Omnibus (GEO).Gene Ontology (GO) Kyoto Encyclopedia Genes Genomes pathway (KEGG) used explore biological functions these common DEGs.Through a series bioinformatic analyses, was established. In additional, external GSE102349 test prognostic value hub mRNAs through Kaplan-Meier method. Results: We successfully constructed NPC, consisting 16 lncRNAs, 6 miRNAs, 3 circRNAs 10 found three (TOP2A, ZWINT, TTK) significantly associated with overall survival time (OS) patients. Conclusion:The revealed this study may help comprehensively elucidate mechanisms driving provide novel candidate biomarkers for evaluating prognosis NPC.

Long non-coding RNAs (lncRNAs) are widely involved in human cancers' progression by regulating tumor cells' various malignant behaviors. MAPKAPK5-AS1 has been recognized as an oncogene colorectal cancer. However, the biological role of hepatocellular carcinoma (HCC) not explored.Quantitative real-time PCR was performed to detect level HCC tissues and cell lines. The effects on growth metastasis were assessed via vitro experiments, including MTT, colony formation, EdU, flow cytometry, transwell assays, nude mice models. western blotting analysis carried out determine epithelial-mesenchymal transition (EMT) markers AKT signaling. interaction between MAPKAPK5-AS1, miR-154-5p, PLAGL2 explored luciferase reporter assay RNA immunoprecipitation. regulatory effect HIF-1α evaluated chromatin immunoprecipitation.MAPKAPK5-AS1 expression significantly elevated HCC, its overexpression associated with clinical features reduced survival. Functionally, knockdown repressed proliferation, mobility, EMT cells induced apoptosis. Ectopic contributed proliferation invasion vitro. Furthermore, silencing suppressed, while enhanced lung vivo. Mechanistically, upregulated PLAG1 like zinc finger 2 (PLAGL2) acting endogenous competing (ceRNA) sponge thereby activating EGFR/AKT Importantly, rescue experiments demonstrated that miR-154-5p/PLAGL2 axis mediated function cells. Interestingly, we found hypoxia-inducible factor 1α (HIF-1α), a transcript factor, could directly bind promoter activate transcription. regulated through form hypoxia-mediated MAPKAPK5-AS1/PLAGL2/HIF-1α signaling loop HCC.Our results reveal suggest be potential novel therapeutic target HCC.

The discovery of long noncoding RNAs (lncRNAs) has improved the understanding development and progression in various cancer subtypes. However, role lncRNAs temozolomide (TMZ) resistance glioblastoma multiforme (GBM) remains largely undefined. In this present study, differential expression was identified between U87 TMZ-resistant (TR) cells. lncRNA XLOC013218 (XLOC) drastically upregulated TR cells associated with poor prognosis glioma. Overexpression XLOC markedly increased TMZ resistance, promoted proliferation, inhibited apoptosis vitro vivo. addition, RNA-seq analysis gain-of-function or loss-of-function studies revealed that PIK3R2 potential target XLOC. Mechanistically, recruited specificity protein 1 (Sp1) transcription factor binding Sp1 to promoters PIK3R2, which elevated both mRNA levels. Finally, PIK3R2-mediated activation PI3K/AKT signaling pathway cell but apoptosis. conclusion, these data highlight vital XLOC/Sp1/PIK3R2/PI3K/AKT axis GBM resistance.
