Bone cancer pain (BCP) impairs patients' quality of life. However, the underlying mechanisms are still unclear. This study investigated role spinal interneuron death using a pharmacological inhibitor ferroptosis in mouse model BCP. Lewis lung carcinoma cells were inoculated into femur, resulting hyperalgesia and spontaneous pain. Biochemical analysis revealed that levels reactive oxygen species malondialdehyde increased, while those superoxide dismutase decreased. Histological showed loss GAD65+ interneurons provided ultrastructural evidence mitochondrial shrinkage. Pharmacologic inhibition ferrostatin-1 (FER-1, 10 mg/kg, intraperitoneal for 20 consecutive days) attenuated ferroptosis-associated iron accumulation lipid peroxidation alleviated Furthermore, FER-1 inhibited pain-associated activation ERK1/2 COX-2 expression prevented GABAergic interneurons.
