Background/Objectives: Male infertility is a significant global health issue, comprising approx. 50% of all cases. Semen cryopreservation, critical component assisted reproductive technologies (ARTs), method commonly used in wide range situations, including gonadotoxic treatments such as radiation or chemotherapy, hazardous occupational exposures, and various medical conditions. Although historically viewed potentially damaging to sperm, recent findings suggest that when performed with appropriate techniques, may fact enhance semen quality by improving the proportion healthy spermatozoa, particularly terms their morphological parameters. The aim this study was evaluate impact cryopreservation on sperm morphology viability, utilizing advanced assessments pre- post-freezing. To evaluate the roles of apoptosis and ferroptosis in cryopreservation-induced damage to sheep sperm, with a focus on assessing effectiveness inhibitors targeting these pathways. Initial analysis compared expression apoptotic marker Cleaved-caspase3 (CL-caspase3) ferroptotic Transferrin receptor (TFRC) between fresh cryopreserved sperm. Abstract Background Freezing-induced sperm damage, often associated with oxidative stress, can result in regulated cell death. Given that stress trigger various forms of death, the prevailing form during cryopreservation remains unknown. Our study aimed to investigate this issue using cashmere goats as a model. Results We found significant increase lyso-phospholipids frozen-thawed suggested ferroptosis. In cryopreservation technology, the choice of cryoprotectant plays a crucial role in cell survival and function.
