Obstructive sleep apnea (OSA) is characterized by intermittent hypoxia/hypercapnia (IHC), affects predominantly obese individuals, and increases atherosclerosis risk. Since we others have implicated gut microbiota metabolites in atherogenesis, dissected their contributions to OSA-induced atherosclerosis. Atherosclerotic lesions were compared between conventionally-reared specific pathogen free (SPF) germ-free (GF) Apoe-/- mice following a high fat cholesterol diet (HFHC), with without IHC conditions. The fecal metabolome profiled using 16S rRNA gene amplicon sequencing untargeted tandem mass spectrometry (LC-MS/MS) respectively. Phenotypic data showed that HFHC significantly increased as regular chow (RC) both aorta pulmonary artery (PA) of SPF mice. exacerbated addition HFHC. Differential abundance analysis identified an enrichment Akkermansiaceae depletion Muribaculaceae (formerly S24-7) family members the HFHC-IHC group. LC-MS/MS dysregulation bile acid profiles taurocholic acid, taurodeoxycholic 12-ketodeoxycholic enriched group, long-chain N-acyl amides, phosphatidylcholines. Interestingly, GF markedly reduced atherosclerotic formation relative under HFHC/IHC In contrast, microbial colonization did not show significant impact on progression PA. summary, this research demonstrated (1) acts cooperatively induce atherosclerosis; (2) modulate induced HFHC/IHC, PA; (3) different analytical methods suggest imbalance bacterial families mediate (4) derived acids, such deoxycholic lithocholic regulate OSA. knowledge obtained provides novel insights into potential therapeutic approaches prevent treat

The pathophysiology of cardiovascular diseases encompasses a complex interplay genetic and environmental risk factors. Even if traditional factors are treated to target, there remains residual risk. This manuscript reviews the potential role gut microbiota in development disease, as target. A systematic search was conducted until 30 October 2024 on PubMed (MEDLINE), using MeSH terms [Gut microbiota] + [Dysbiosis] [Cardiovascular] [TMAO] [bile acids] [short-chain fatty acids]. term dysbiosis implies changes equilibrium, with modifications composition functionality series additional factors: reduced diversity uniformity microorganisms; short-chain acid-producing bacteria; increased permeability; release metabolites, such trimethylamine N-oxide, betaine, phenylalanine, tryptophan-kynurenine, phenylacetylglutamine, lipopolysaccharides; secondary bile acid excretion, leading inflammation, oxidative stress, endothelial dysfunction facilitating onset pathological conditions, including obesity, hypertension, diabetes, atherosclerosis, heart failure. Attempts restore balance through different interventions, mainly diet, have been shown positively affect individual components metabolites reduce disease. In addition, probiotics prebiotics potentially useful. Fecal transplantation is promising therapy.

Atherosclerotic cardiovascular diseases (ASCVDs) remain a leading cause of mortality, with early cholesterol control being pivotal in mitigating long-term risk. Recent findings suggest that intermittent hyperlipidemia, characterized by oscillatory exposure, uniquely accelerates atherosclerosis compared to continuous high-fat diets. This review synthesizes emerging evidence on hyperlipidemia's impact atherogenesis, emphasizing macrophage dysfunction, autophagy impairment, and efferocytosis deficits. We also discuss critical gaps, including sex-specific differences, gut-microbiota interactions, the influence maternal hypercholesterolemia. Notably, recent insights into IL-1β-dependent neutrophil reprogramming under diets reveal novel inflammatory mechanisms driving plaque destabilization. Addressing these gaps will advance our understanding atherogenesis guide development innovative prevention strategies therapeutic interventions.

Abstract Excessive accumulation of reactive oxygen species (ROS) and dysbiosis gut microbiota are pivotal etiological factors in the pathogenesis ulcerative colitis (UC) its associated intestinal extraintestinal manifestations (e.g., microthrombosis, anxiety, depression symptoms). This investigation presents a multifunctional inulin complex (PB/NKase@Inulin gel) incorporating Prussian blue nanozymes (PB NZs) thrombolytic agent nattokinase (NKase) for therapeutic management dextran sulfate sodium (DSS)‐induced UC complications. Following oral administration, PB/NKase@Inulin gel, characterized by prolonged retention PB NZs NKase at inflamed colonic sites, can facilitate continuously ROS scavenging, attenuate oxidative stress damage, effectively reduce pro‐inflammatory cytokine levels. Importantly, gel not only robustly inhibit inflammatory microthrombosis formation but also lyses thrombi due to potent properties both vitro vivo. Furthermore, is able modulate homeostasis alleviate multiple stresses responses (including anxiety depression) mouse model via microbiota‐gut‐brain (MGB) axis interactions. Overall, offers an innovative paradigm comprehensive interventions DSS‐induced multifaceted
