Abstract Cholangiocarcinoma (CCA) includes a cluster of highly heterogeneous biliary malignant tumours that can arise at any point the tree. Their incidence is increasing globally, currently accounting for ~15% all primary liver cancers and ~3% gastrointestinal malignancies. The silent presentation these combined with their aggressive nature refractoriness to chemotherapy contribute alarming mortality, representing ~2% cancer-related deaths worldwide yearly. current diagnosis CCA by non-invasive approaches not accurate enough, histological confirmation necessary. Furthermore, high heterogeneity CCAs genomic, epigenetic molecular levels severely compromises efficacy available therapies. In past decade, efforts have been made understand complexity develop new diagnostic tools therapies might help improve patient outcomes. this expert Consensus Statement, which endorsed European Network Study Cholangiocarcinoma, we aim summarize critically discuss latest advances in CCA, mostly focusing on classification, cells origin, genetic abnormalities, alterations, biomarker discovery treatments. horizon next decade from 2020 onwards highlighted.

Tumor-associated macrophages (TAMs) represent one of the main tumor-infiltrating immune cell types and are generally categorized into either two functionally contrasting subtypes, namely classical activated M1 alternatively M2 macrophages. The former typically exerts anti-tumor functions, including directly mediate cytotoxicity antibody-dependent cell-mediated (ADCC) to kill tumor cells; latter can promote occurrence metastasis cells, inhibit T response, angiogenesis, lead progression. Both have high degree plasticity thus be converted each other upon microenvironment changes or therapeutic interventions. As relationship between TAMs malignant tumors becoming clearer, become a promising target for developing new cancer treatment. In this review, we summarize origin TAMs, interaction with microenvironment, up-to-date treatment strategies targeting TAMs.

Abstract Circulating tumor cells (CTCs) are that have sloughed off the primary and extravasate into circulate in blood. Understanding of metastatic cascade CTCs has tremendous potential for identification targets against cancer metastasis. Detecting these very rare among massive blood is challenging. However, emerging technologies detection profoundly contributed to deepening investigation biology facilitated their clinical application. Current summarized herein, together with advantages disadvantages. The usually dependent on molecular markers, epithelial cell adhesion molecule being most widely used, although markers vary between different types cancer.
