To understand the dynamic interplay between human microbiome and host during health disease, we analyzed microbial composition, temporal dynamics, associations with multi-omics, immune, clinical markers of microbiomes from four body sites in 86 participants over 6 years. We found that stability individuality are body-site specific heavily influenced by host. The stool oral more stable than skin nasal microbiomes, possibly due to their interaction environment. identify individual-specific commonly shared bacterial taxa, individualized taxa showing greater stability. Interestingly, dynamics correlate across sites, suggesting systemic host-microbial-environment interactions. Notably, insulin-resistant individuals show altered among microbiome, molecular markers, features, disrupted metabolic disease. Our study offers comprehensive views multi-site relationship

The levels of the thousands metabolites in human plasma metabolome are strongly influenced by an individual's genetics and composition their diet gut microbiome. Here, assessing 1,183 1,368 extensively phenotyped individuals from Lifelines DEEP Genome Netherlands cohorts, we quantified proportion inter-individual variation explained different factors, characterizing 610, 85 38 as dominantly associated with diet, microbiome genetics, respectively. Moreover, a quality score derived metabolite was significantly quality, assessed detailed food frequency questionnaire. Through Mendelian randomization mediation analyses, revealed putative causal relationships between metabolites. For example, analyses support potential effect Eubacterium rectale decreasing hydrogen sulfite-a toxin that affects cardiovascular function. Lastly, based on analysis 311 at two time points separated 4 years, observed positive correlation stability amount variance could be our analysis. Altogether, characterization factors explain can help design approaches for modulating or to shape healthy metabolome.

Bifidobacteria are among the earliest and most abundant bacterial colonizers of neonatal gut in many mammals, where they elicit purported host health benefits. While early life-associated dynamics diversity, as well metabolic beneficial activities, Bifidobacterium species have been studied, functional contributions bifidobacteria to well-being adults remain less explored. In this opinion piece, we discuss current knowledge regarding relevance endogenous associated with adulthood. We identify gaps opportunities for microbiota enrichment rationally selected strains more adapted adult host. propose that future studies area will help us better understand ecological, metabolic, roles played by ecosystem across various ages.

Oxidative stress (OS) is a key pathophysiological mechanism in Crohn's disease (CD). OS-related genes can be affected by environmental factors, intestinal inflammation, gut microbiota, and epigenetic changes. However, the role of OS as potential CD etiological factor or triggering unknown, differentially expressed either cause subsequent change inflammation. Herein, we used multi-omics summary data-based Mendelian randomization (SMR) approach to identify putative causal effects underlying mechanisms CD.

Antibiotics (ABX) compromise the efficacy of programmed cell death protein 1 (PD-1) blockade in cancer patients, but mechanisms underlying their immunosuppressive effects remain unknown. By inducing down-regulation mucosal addressin adhesion molecule (MAdCAM-1) ileum, post-ABX gut recolonization by Enterocloster species drove emigration enterotropic α4β7+CD4+ regulatory T 17 cells into tumor. These deleterious ABX were mimicked oral gavage species, genetic deficiency, or antibody-mediated neutralization MAdCAM-1 and its receptor, α4β7 integrin. contrast, fecal microbiota transplantation interleukin-17A prevented ABX-induced immunosuppression. In independent lung, kidney, bladder patient cohorts, low serum levels soluble had a negative prognostic impact. Thus, MAdCAM-1-α4β7 axis constitutes an actionable immune checkpoint immunosurveillance.

Abstract Major depressive disorder (MDD), bipolar (BD) and schizophrenia-spectrum disorders (SSD) are heterogeneous psychiatric disorders, which place significant burden on patient's well-being global health. Disruptions in the gut-microbiome may play a role these disorders. This review presents current data composition of human gastrointestinal microbiota, its interaction mechanisms gut–brain axis MDD, BD SSD. Diversity metrics microbial relative abundance differed across studies. More studies reported inconsistent findings ( n = 7) or no differences 8) than who lower α -diversity 5). The most consistent were higher abundances genera Streptococcus , Lactobacillus Eggerthella butyrate producing Faecalibacterium patients with All three increased associated symptom severity. Confounders, such as medication use life style have not been accounted for. So far, results probiotics trials inconsistent. Most traditional widely used (consisting Bifidobacterium spp. spp.) safe, however, they do correct potential microbiota disbalances Findings prebiotics faecal transplantation (FMT) too limited to draw definitive conclusions. Disease-specific pro/prebiotic treatment even FMT could be auspicious interventions for prevention therapy should investigated future trials.
